E. coli BacPowerTM Guaranteed Package Quotation Form Instructions Please answer all of the following questions and send the completed form to protein@genscript.com if you choose the E. coli BacPowerTM Guaranteed Package. Our service representative will contact you with a quote shortly. Customer Information If you have registered an account with GenScript, you can just identify yourself by giving us your name and email address or Account No. Name: Account No.: Phone: Organization: Shipping address: (Necessary to determine the shipping cost) Email address: Project Information Nature of inquiry: Order initiation: Quote for ordering Immediately For pricing estimation Within three months For grant application Beyond three months Please read the following terms and conditions for the E. coli BacPowerTM Guaranteed Package: Terms and Conditions: Protein sequences are subject to pre-project analysis. Exclusions are applicable for membrane proteins and toxic proteins. For eligible proteins, a minimum of 3 mg soluble protein with >75% purity will be delivered to customers. The protein service will be free of charge if the expression level is too low to eventually obtain 3 mg of purified protein after Genscript’s evaluation. To increase the expression level of your protein, optimized genes are recommended using our OptimumGeneTM technology (note: gene synthesis is at additional cost). Up to 3 fusion tag(s) may be tested to increase the expression level and solubility of the protein and applied for protein production and purification. Fusion tags may be made removable by introducing optional protease cleavage site between the tag and the target protein (note: Tag removal service is available at extra cost). Expressed insoluble proteins will be subject to refolding using our FoldArt TM technology at no additional fee. Protein Expression & Purification Protein name: Species: MW: Protein sequence (crucial for evaluation) or accession number: Note: Please enter the protein sequence if mutants are included in your protein sequence. Gene synthesis with codon optimization (For Human/Mouse/Rat ORF genes exist in NCBI database, starting from $0.29/bp; for other genes, starting from $0.35/bp) (Please enter your gene sequence: ) Starting material: Gene synthesis according to natural gene sequence (it is strongly recommended that synthesized gene with optimized codon be used for protein expression) (Please enter your gene sequence: ) Your own template DNA (it is strongly recommended that synthesized gene with optimized codon be used for protein expression) (Please attach vector map (e.g. NTI file), and enter your vector name: , and gene sequence: Cytoplasmic protein Periplasmic protein Secreted protein IgG antibody Membrane-bound protein Toxic to E. coli Protein properties: Tag position: Note: If the target protein is membrane-bound or toxic to the bacterial host, please quote with E. coli customized service. Do you want GenScript to add the tag(s) at the N-terminus? Yes No, I have designated place(s) to add the tag(s) (Please specify): Note: If internal tag(s) is/are involved, please designate the position number(s) that corresponds to the sequence of which the accession number you have indicated above. Please select up to three tags to facilitate the expression and purification of your target protein: Tag selection: Introducing Protease cleavage site: Protein characterization: 6xHis GST Other (please specify): MBP Trx Sumo Note: A customer selected tag or small tag (e.g. -6xHis) will be the first choice for protein production. GenScript reserves the right to choose a best tag in order to guarantee the target protein expression. Do you want GenScript to add a protease cleavage site between the tags and the target protein? Yes No If YES, please specify the protease cleavage site you want to add: GenScript’s choice Enterokinase PreScission rTev Thrombin Note: After protease cleavage, an extra 4-6 aa will be left of C-terminal tag and <2 aa of N-terminal tag; therefore, we DO NOT recommend the introduction of protease cleavage site between the target protein and the C-terminal tag. SDS-PAGE & Western blot SEC ELISA HPLC MS MALDI-TOF N-terminal sequence Note: SDS-PAGE & Western blot is FREE of charge. Other comments TF
