PRODUCT INFORMATION
Description
Recombinant Human MCP-4 produced in E.Coli is a non-glycosylated, Polypeptide
chain containing 75 amino acids and having a molecular mass of 8,510 Dalton.
Human MCP-4 is a CC chemokine recently cloned from a human fetal and a human
heart cDNA library. Human MCP-4 cDNA encodes a 98 amino acid residue precursor
protein with a 23 amino acid residue hydrophobic signal peptide that is cleaved to yield
an 8kDa, 75 aa mature MCP-4. Mature MCP-4 lacks any potential N-glycosylation sites
and shares a pyroglutamate proline motif with other human MCP proteins. Human
MCP-4 is most homologous to MCP-1, 3 and Eotaxin, exhibiting approximately 65 66% amino acid sequence identity. MCP-4 mRNA is expressed by a number of
activated cell types, including endothelial cells, macrophages, bronchial epithelium and
type II alveolar cells, and perhaps lymphocytes. In addition, it has been reported to be
chemotactic for CD4+ and CD8+ T cells, with an activity almost equivalent to that of
MCP-3. The bioactivities of MCP-4 is most likely mediated by the CC chemokine
receptors CCR-2 and CCR-3, both of which have been shown to bind MCP-4.

Quantitation
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm.
2. RP-HPLC-Analysis, using a standard solution of MCP-4 as a Reference Standard.
Specific Activity
The specific activity as determined by the ability of MCP-4 to chemoattaract human
monocytes at 10-100ng/ml.
Storage
Lyophilized rHuMCP-4 although stable at room temperature for 3 weeks, should be
stored desiccated below -18 °C. Upon reconstitution rHuMCP-4 should be stored at
4 °C between 2-7 days and for future use below -18 °C. For long term storage it is
recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw
cycles.
Formulation
The protein was lyophilized from a concentrated (1 mg/ml) sterile solution containing no
additives.
Reconstitution
It is recommended to reconstitute the lyophilized rHuMCP-4 in sterile 18 MΩ-cm H2O
not less than 100 μg/ml, which can then be further diluted to other aqueous solutions.

Dimers and aggregates
Less than 1% as determined by silver-stained SDS-PAGE gel analysis.
Sequence analysis
The sequence of the first five N-terminal amino acids was determined and was found to
be Gln-Pro-Asp-Ala-Leu.
