Description
Hsp27, also known as the Estrogen-Regulated 24K protein, and hsp 28 is a member of
the mammalian small heat shock protein family. Hsp27 is expressed constitutively in
many tissues and its expression is increased to high levels after various types of stress
including elevated temperatures, toxic metals, drugs and oxidants. Also, Hsp27 is
phosphorylated in vivo on three phosphorylation sites (Ser15, Ser78 and Ser82) by
protein kinases including MAPKAP kinase 2 and the stress-activated protein kinase
SAPK2 (p38). Recombinant HSP-27 produced in E.Coli is a single, non-glycosylated
polypeptide chain containing 205 amino acids and having a molecular mass of 22.7
kDa. Recombinant Human Hsp27 was overexpressed in E. coli and purified by
conventional chromatography.

Quantitation
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm.
2. Analysis by RP-HPLC, using a calibrated solution of HSP-27 as a Reference
Standard.
Specific Activity
Immunoreactivity is confirmed by reaction with monoclonal mouse antibodies against
HSP 25/27.
Storage
Lyophilized rHuHSP-27 although stable at room temperature for 3 weeks, should be
stored desiccated below -18 °C. Upon reconstitution rHuHSP-27 should be stored at
4 °C between 2-7 days and for future use below -18 °C. For long term storage it is
recommended to add a carrier protein (0.1% HSA or BSA). Please prevent
freeze-thaw cycles.
Formulation
The protein was lyophilized from a concentrated (1 mg/ml) solution containing 20 mM
tris/acetate pH 7.6, 10 mM sodium chloride, 0.1 mM EDTA, 0.1 mM PMSF and 15 mM
A-mercaptoethanol.
Reconstitution
It is recommended to reconstitute the lyophilized rHuHSP-27 in sterile 18 MΩ-cm H2O
not less than 100 μg/ml, which can then be further diluted to other aqueous solutions.
