Eotaxin (murine) PRODUCT INFORMATION Description Eotaxin (EOT) is a member of the C-C, or β chemokine class. Eotaxin was originally isolated from bronchoalveolar lavage fluid of guinea pigs sensitized by aerosol challenge with ovalbumin. EOT is an approximately 8.4 kDa polypeptide of 74 amino acids. The precursor form of eotaxin consists of 97 amino acids. To generate the mature eotaxin (74 amino acids), a 23 amino-terminal amino acid signal peptide is cleaved from the precursor. EOT is chemotactic for eosinophils, but not mononuclear cells or neutrophils. Human EOT shows approximately 60% amino acid homology to mouse and guinea pig eotoxin. Eotaxin also shows high identity with MCP-1, 2, and 3. Recombinant Murine Eotaxin produced in E. coli is a single, non-glycosylated polypeptide chain containing 74 amino acids and having a molecular mass of 8,403.2 Dalton. The rmEotaxin is purified by proprietary chromatographic techniques. Cat. No.: Z00302 Size: 1 mg or 2 ug or 10 ug Source: Escherichia coli. MW: 8,403.2 Dalton Endotoxin Level: Less than 0.1 ng/μg (IEU/μg) of rmEotaxin. Dimers and aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis. Version: 08/24/2006 Purity Greater than 98.0% as determined by: (a) Analysis by RP-HPLC. (b) Anion-exchange FPLC. (c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel Quantitation Protein quantitation was carried out by two independent methods: 1. UV spectroscopy at 280 nm. 2. Analysis by RP-HPLC, using a standard solution of mEotaxin as a Reference Standard. Specific Activity The Biological activity is determined by its ability to induce chemotaxis purified eosinophils at a concentration of 100-1,000 ng/ml. Storage Lyophilized rmEotaxin although stable at room temperature for 3 weeks, should be stored desiccated below -18 °C. Upon reconstitution rmEotaxin should be stored at 4 °C between 2-7 days and for future use below -18 °C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles. Formulation Lyophilized from a concentrated (1 mg/ml) solution in water containing no additives. Reconstitution It is recommended to reconstitute the lyophilized rmEotaxin in sterile 18 MΩ-cm H2O not less than 100 μg/ml, which can then be further diluted to other aqueous solutions.
