Interleukin (IL)-11, human PRODUCT INFORMATION Description IL-11 is a thrombopoietic growth factor that directly stimulates the proliferation of hematopoietic stem cells and megakaryocyte progenitor cells and induces megakaryocyte maturation resulting in increased platelet production. IL-11 is a multifunctional cytokine that has pleiotropic effects on a variety of tissue including the bone marrow and intestinal mucosa. IL-11 also stimulates proliferation of primitive stem cells and multipotential and committed progenitor cells from various sources including cord blood, bone marrow, and peripheral blood in different culture systems and increases the commitment of primitive stem cells into the progenitor cell compartment. IL-11 sustains hematopoiesis resulting in an increased number of megakaryocytes and erythropoiesis, lymphopoiesis, and myelopoiesis. IL-11 acts synergistically with other early and late acting growth factors that stimulate various stages and lineages of hematopoiesis. Cat. No.: Z00375 Size: 1 mg or 50 ug or 10 ug Source: P. Pichia. Endotoxin Level: Less than 2 ng/mg (2 IEU/μg) determined by LAL test. MW: 19.2 KD+/-10% determined by reduced SDS-PAGE. Version: 03/21/2007 UV Scan The maximal absorption wave is 278+/-3 nm. Purity Greater than 95% as determined by: (a) Analysis by SEC-HPLC. (b) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel. Residual DNA: less than 2 ng/mg analysis by solid phase blot. Residual host cell protein: less than 0.1% analysis by ELISA. Specific Activity The specific activity as determined was found to be greater than 8.0×10 6 IU/mg. Storage Lyophilized rHuIL-11 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution rHuIL-11 should be stored at 4°C between 2-7 days and for future use below -18°C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles. Formulation The protein was lyophilized from a concentrated (1 mg/ml) solution with no additives. Reconstitution It is recommended to reconstitute the lyophilized rHuIL-11 in sterile 18 MΩ-cm H2O not less than 100 μg/ml, which can then be further diluted to other aqueous solutions. Dimers and aggregates Residual DNA: less than 2 ng/mg analysis by solid phase blot. Residual host cell protein: less than 0.1% analysis by ELISA. Sequence analysis The sequence of the first fifteen N-terminal amino acids was determined and was found to be Gly-Pro-Pro-Pro-Gly-Pro-Pro-Arg-Val-Ser-Pro-Asp-Pro-Arg-Ala. ** For non-clinical research use only. ** Tel: 732-885-9188 Fax: 732-210-0262 E-Mail: info@genscript.com www.genscript.com
