Application for use of the Small Molecule Screening Facility Revised by NRP 2010 User Information: Name E-mail Principal Investigator Email: Telephone # Lab Name (Name of PI) Cancer Center Member? Yes No ( circle one) Billing Information: Billing Contact Name: Billing Contact Phone: Billing Contact Email: Grant to charge: The SMSF will obtain funding information from your billing contact and supply a copy of each invoice. By applying for SMSF services, I understand that all activities performed at the SMSF incur charges as outlined below, and a fund account number will be provided within 1 week of receiving an invoice. I understand that if a fund account number is not provided within 1 week, that additional billable hours may be generated to cover administrative time required to obtain a fund account number. Signature of Principal Investigator: ________________________________________ Note: A copy will be sent to your billing contact for their reference. SMSF Rates and Policies: All users of the SMSF and the MCCF pay user fees to offset the costs of staffing, instrument maintenance, and operational expenses. Initial project consulting is provided free of charge (not to exceed 2 hours). User participation is encouraged at all times. Hourly Staff Time for Assisted Facility and Equipment Use, Chemistry and Screening Consulting, reporting and data analysis: UW Investigator: Off Campus Investigator: $150.00 per hour* ** $300.00 per hour** Hourly Staff Time for User Assisted Facility and Equipment Use (UW Member Only)***: UW Investigator $75 per hour* ** Application for use of the Small Molecule Screening Facility Revised by NRP 2010 *Members of the Carbone Cancer Center receive 25% subsidy of all project costs including consumables and reagent costs. **Users pay for all consumables and reagents required for their project. ***Large liquid handlers, HPLC, chemical libraries, and RNAi libraries are restricted and must be handled/operated by facility staff Walk-up: Access to facility instruments without the assistance of staff is available for some instruments. Charges are applied for amount of time reserved. A complete listing of instruments is available on our website: http://www.hts.wisc.edu/scheduling.php Compound Access Fees: To offset the costs associated with maintaining the compound libraries, a small fee is charged per library: KBA01-08: $200 Chembridge: $500 Maybridge: $500 Chemdiv: $600 Life Chemicals $700 Life Chemicals 2: $700 Charges are based upon the size of the library. Charge is assessed 1X per project. RNAi Access Fees: To offset the costs associated with maintaining the RNAi libraries, a $2500 is charged for use of the libraries. Cytotoxicity and Cell Proliferation Assay Costs per Compound: UW Investigator: $175 per compound for preliminary 2 dose test in a cell panel including 2 of the following cell lines: Du145, MCF7, HT-29, Hep3B, A549, SF-268, MB-MDA-231.* An additional $175 per compound for full IC50 in 1-7 cell lines listed above. An additional $350 will be charged for the controls per plate of 9 compounds ($175 preliminary assay/$175 full screen). * To be sure we cover the costs to maintain the cells, there is a 9 compound/1 control minimum, if less compounds are tested the minimum cost is $1750.* For additional cell lines, $50 per compound per cell line, again 9 cpd/1 control minimum or $500. Application for use of the Small Molecule Screening Facility Revised by NRP 2010 *Carbone Cancer Center members receive a 25% subsidy of all project costs including consumables and reagent costs. Non-UW Investigators: $400 per compound for preliminary 2 dose test in a cell panel including: Du145, MCF7, HT-29, Hep3B, A549, SF-268, MB-MDA-231. An additional $400 per compound for full IC50 in 1- 7 cell lines listed above. An additional $600 will be charged for the control per plate of 9 compounds tested in preliminary and full IC50 screens. To be sure we cover the costs to maintain the cells, there is a 9 compound/1 control minimum, if less compounds are tested the minimum cost is $4000.00.* For additional cell lines, $125 per additional cpd/cell line, 9 cpd/1 control minimum or $1000. Service Information: 1. Briefly describe your intended use of the SMSF (HTS, cytotoxicity assay, etc.). 2. For HTS, RNAi, and Mechanism assays please summarize your project: 3. Please briefly describe your protocol in general terms. (Eg 1. Add 30 L labeled proteins in buffer to 384 well plates. 2. Add 1 L 1mM compound to each well. 3. Incubate at room temp. 1hr. 4. Read in plate reader using LANCE protocol.) (N/A if for Walk-up Instruments) 4. Please list materials needed at the screening facility for your project (plates, filter wavelenths, etc.) –note all materials costs will be billed to the investigator.
