SOP Number: CP034.00 Title: Preparation of Blood, Tissue, and Urine Sample Containers Revision No: Replaces: 00 N/A Author: Tatiana Zanganeh I Date in effect: Page: 7/8/2010 Page 1 of 4 MAL Director: Dr. Peggy Borum FSHN Chair: Dr. Neil Shay PURPOSE Tissue and blood sample containers are used to transport and store samples so they can be used for analysis and/or assays. II SCOPE This procedure outlines the preparation of containers for storing tissue and blood samples. During non-survival surgery, organs may be collected for analysis. It is important to correctly label these containers so they can be properly organized and used later. Assigning each piglet a ‘piglet identification number’ is an easy way to keep track of samples. III RESPONSIBILITIES It is the responsibility of Metabolic Assessment Laboratory personnel to follow this procedure. It is the responsibility of supervisory personnel to ensure compliance with this procedure and to train employees and students responsible for performing this procedure. Students will report accidents to the principal investigators immediately. IV REFERENCES N/A V REAGENTS AND MATERIALS V.A. V.B. V.C. V.D. V.E. V.F. V.G. V.H. V.I. V.J. Members of the MAL have been trained extensively in the procedures described in this SOP. VIII DEFINITIONS VIII.A. Standard Operating Procedure (SOP) – Standard Operating Procedure is a document that provides instructions for completing a specific task in the lab. VIII.B. Metabolic Assessment Laboratory (MAL) – The Metabolic Assessment Laboratory is the laboratory that will use this SOP. VIII.C. Piglet Identification (ID)—The Piglet ID is a unique, alphabetical and numerical label that identifies the piglet and the samples taken from that piglet. IX PROCEDURE IX.A. IX.B. IX.C. Preparation of the tissue collection weigh boats. IX.A.1. Refer to Attachment I for number of weigh boats needed. IX.A.2. Label the outer edge of the plastic weigh boat in the following manner with a fine tip black permanent marker: IX.A.2.a Piglet ID. IX.A.2.b Name of organ. Preparation of the tissue collection vials. IX.B.1. Refer to Attachment I for number of vial labels needed. IX.B.2. Using a fine tip black marker, label the top of the plastic vial with the following: IX.B.2.a. Piglet ID. IX.B.2.b. Name of organ. IX.B.3. Tape a paper label to the side of the vial making sure to cover the paper label and encircle the vial completely with tape to ensure a water tight seal with the following information: IX.B.3.a. Piglet ID. IX.B.3.b. Name of organ. IX.B.3.c. Date of non-survival surgery. IX.B.4. Be sure to evenly center the label on the vial and wrap the tape around completely. Preparation of the blood collection Vacutainer tubes. IX.C.1. Tape a paper label to the side of the Vacutainer tube perpendicular to the length of the Vacutainer tube, using the same technique as above with the following information: IX.C.1.a. Piglet ID. IX.C.1.b. “Central Blood” or “Portal Blood”. IX.C.1.c. Date of non-survival surgery. IX.C.2. Repeat the procedure with the correct number of Central Blood and Portal Blood Vacutainer tubes. Refer to the following information to tape paper labels to plastic test tubes perpendicular to the length of the test tube using the technique mentioned above. IX.D.1.a. Piglet ID. IX.D.1.b. Name of sample. IX.D.1.c. Date of non-survival surgery. IX.D.2. Repeat the procedure with the following number of blood analysis tubes per piglet: IX.D.2.a. 14 Washed RBC. IX.D.2.b. 40 Plasma. IX.D.2.c. 1 Blood for Hgb. IX.D.2.d. 1 Portal for Hgb. IX.D.2.e. 3 Portal Plasma. IX.D.2.f. 3 Portal Washed RBC. Preparation of the 20mL scintillation vial for urine collection. IX.E.1. Label 20 mL scintillation vial caps with a fine tip permanent marker the following information: IX.E.1.a. Piglet ID. IX.E.1.b. Urine. IX.E.1.c. Date of urine sample. IX.E.1.d. Time of hourly urine draw. IX.E.2. Tape a paper label using the technique mentioned above to the side of the plastic vial as follows: IX.E.2.a. Piglet ID. IX.E.2.b. Urine. IX.E.2.c. Date of urine sample. IX.E.2.d. Time of hourly urine draw. IX.E.3. A separate vial needs to be prepared for every two samples of urine drawn on the half hour (one vial every hour).
