Gel Electrophoresis Results: After different DNA fragments and plasmids have been separated by gel electrophoresis, the gel is stained to show bands that indicate the location of each kind of fragments and plasmid: 1) What is the approximate size of the fragments (A – E)? What is the order of the fragments, from smallest to largest? 2) Look at the gray boxes: Compare the lanes that have linear fragments with the lanes that have plasmids. Is there a difference in the shape of the bands between these two DNA forms? In which lane do you expect to find the rfp gene and the ampR gene in the gel photograph? In the geK+ and the geA+ lanes, do you see evidence of complete digestion? 3) These fragments were created from the digestion of pKAN and pARA with BamHI and HindIII. Draw 3 possible recombinant plasmids that you could make by ligating the digested fragments of the pKAN-R and the pARA plasmids.
