Screening of germplasm for drought tolerance using different drought related traits.
Crossing of identifies drought resistant germplasm/line(s) with susceptible elite rice variety.
Identification of molecular markers tightly linked to trait(s) related to drought resistance.
Isolation of mRNA from control, and stressed samples of rice.
cDNA synthesis from the isolated mRNA’s.
Database search and retrieval of information for genes expressed during stress.
Primer designing and PCR amplification of the reported stress inducible genes.
Sequencing of the cloned genes and database search for gene identification/confirmation.
Complete nucleotide sequencing and categorization of the cloned genes.
Current Knowledge about Genomics Lab.
1. Actively involved in Genome Mapping Lab. and green house studies for Rice and
cotton.
2. Very much familiar with the techniques routinely used in Lab. like, total genomic
DNA isolation, Polymerase Chain Reaction (PCR) amplification,
Electrophoresis, Gel documentation systems,
3. Random Amplified Polymorph DNA (RAPD), Simple Sequence Repeat (SSR),
and Restriction Fragment Length Polymorphism RFLP markers Identification.
4. Elution of DNA bands from agarose gel, recovery from the agarose gel, ligation
in TA cloning vector, E.coli transformation, plasmid isolation and restriction.
5. Some knowledge about the SCAR markers.
6. Differential Display: Total RNA isolation, cDNA synthesis, and DDRTPCR.
7. Use of Mapmaker and QTL Cartography.
