Endometritis is one of the most common factor for reproductive failure in farm animals.
Indiscriminate and prolong use of antimicrobial drugs causes resistance of bacteria. Thus,
identification and determination of drugs sensitivity of causative organism has become very
important for effective therapy for gynecological infection. It limits to some extent of drug
resistance strain.
Therefore present investigation was carried out to study conception rate by proper use of
antibiotic which derived from antibiotic sensitivity test.
Material and Method:The experiment was conducted on 87 cross bred cows from village “Charada”. These animals
were selected on the basis of history of repear breeding due to endometritis. Majority of animals
were having mucopurulent discharge at the time of oesrus, on rectal palpation examination by
local veterinary of animal health clinic”Charada”. Uterine sample were collected aseptically and
sensitivity test for the bacteria was performed in BAIF DUDHSAGAR INVESTIGATION
LABORATORY at Vihar Chilling center, Vihar, Ta:- Mansa, Dist:- Gandhinagar.in laboratory
first Recording of sample number and other details. Uterine discharge samples received
in the laboratory and from field veterinarian are registered in the laboratory
Drug sensitivity test
In drug sensitivity test first Incubate sample at 370C for 24hrs in Brain Heart Infusion
broth for better bacterial growth. After incubation Load Sample on Mulear Hilton
Agar Plate and put different antibiotic discs keep it in incubator for 37 0C for 24hrs.
After incubation Prepare smear from the bacterial growth and stained by Giemsa
Stain for identification of Bacteria (Gram +ve or Gram –ve) and observe the zone of
growth inhibition, if the drug is effective. Diameter of inhibition zone is measured and
compared with different antibiotics to interpret the result.
Zone of inhibition around the disc :---: culture sensitive to antibiotics
No Zone of inhibition around the disc :---: culture resistant to antibiotics
The results are communicated to veterinarian for treatment through cow/buffalo owner the
owner get treated his cow according to the report given by D.S.Lab.
In short, the antibiotic sensitivity test is commonly carried out in laboratory so to suggest the
proper treatment to the clinician. In this test the pathogenic bacteria are isolated in from clinical
sample and the same bacteria is treated with the various antibiotic disc to find out the efficiency
of the antibiotics on bacterial culture.
The organism isolated are to be tested for in-vitro sensitivity, here chemotherapeutic agent are
used which are commonly used in field.

The animals are being treated with the drugs selected on the basis of sensitivity report.The
drugs are being given intra uterine for 5 to 6 doses. The animals were re examined in
subsequent estrus and inseminated with frozen semen. The response to the treatment will be
assessed on the basis of pregnancy diagnosis between 60 to 90 days.
The data thus obtained will be statistically analyzed accordingly.

Summary:Out of 87 cross bred cows( suffering from endometritis were treated by Different drugs based on
antibiotic sensitivity test) 50 animals were pregnant means 57.47% animal were pregnant.
Cefriaxone is most sensitivity drug in endometritis. Conception rate of animal which treated by
Ceftriaxone was 65.38%.
