Sample submission guidelines: DNA sequencing can be submitted to the Genomics Core in three formats: 1) DNA template and sequencing primer in separate tubes: Per sequencing reaction provide 10 ul template (template @ 200 ng/ul) and 10 ul primer (primer @ 1 pmol/ul). 2) DNA template and sequencing primer premixed: Per sequencing reaction provide 11 ul of sample containing 200 ng of DNA per 1 kb of template and 10 pmol of primer for plasmid templates and 35-40 ng of DNA and 5 pmol of primer for PCR products. 3) DNA template and sequencing primer premixed in a 96-well format: Per sequencing reaction provide 5 ul of sample containing 100 ng of DNA per 1 kb of template and 5 pmol of primer for plasmid templates and 10 ng of DNA per 100 bp of template and 5 pmol of primer for PCR products. Important Note: the amounts give above are guidelines that function as starting points for optimizing your specific sequencing reactions. Many clients of the Genomics Core have come up with their own amounts and concentrations that work based on their specific templates / DNA isolation methods / DNA quantification methods. If you are a new customer and are uncertain what will work for you please contact the Core manager and we will run some free test samples. Sample collection times and drop-off points on the CCF campus: Samples are collected once a day between 9:00 a.m and 9:30 a.m. For your convenience there are a total of three sample drop-off areas on campus: at LRI one is in the small office corridor in the middle of the west NC1 hallway and one is in the 5th floor NB hallway, opposite the Chairman's office. For those who would like to drop off their samples in a refrigerated spot there is a Genomics Core refrigerator in the NN building: past conference room NC1-202 through the door marked "Area NN". For clients without CCF access there is a refrigerated drop-off point in the ND shipping and delivery area (small fridge on table).
