Bio 415 Final exam. Student Demonstration.
Hi Folks, here are several suggested Rubrics for the biotech students to follow. If you
need to modify them then yes do that. If you have questions please let me know.
The grading is by % and is “Holistic” Meaning give them an overall score on how well
you think they did. Each part will be given a value between 0% and 100%.
A 100% is perfect with absolutely no problems and no hesitation.
A 95% is a did everything correct but had some hesitation and one slip up.
A 90% is a did one or two small things wrong but overall is correct.
We are going to assign one proctor per student. We were thinking of having proctors
stay at particular stations, however the logistics of this became very complicated quickly.
In having one proctor per student we can compartmentalize equipment and proctors so
that students do not overlap.
The students will only know when they will be tested. They will not know who their
proctor is or what their particular test is. Though, they have all been told what the
possibilities are. I would like these little tests to be as hands on as possible. Have the
students do the task rather than describe them. Remember this is the show what you can
do part of the exam.
I will briefly try and describe the tasks and give a grading rubric for them below.
Materials may be set up in multiple rooms. Please have the student change rooms at
least once.
Microscope:
The student will be asked to place a slide in the microscope and successfully bring it into
focus under 400x total power. (Not Oil Immersion, Unless PAT did you go over oil
immersion?)
The students should start with a microscope that has all of its settings out of whack.
Students should have the SOP near. It is ok if the student looks at their notes or the SOP.
Student will safely and in the correct manner place a slide onto the Microscope and bring
it into focus.
Big mistakes:
Student only uses one eye to look into microscope.
Student cannot safely focus slide.
Student cannot focus slide
Small mistakes:
Student uses the course focus when transferring in-between oculars
Student does not know parts of microscope when asked
Student does not know how to determine the power of a microscope
Student does not properly bless the microscope before beginning.
Micropipette
Student will be asked to pipette out between 15 and 850ul from a stock solution into a
micropipette. Students will be given a choice between sterile and non-sterile tips (both
yellow and blue) and a set of pippetmen. (20,200 and 1000). The SOP will also be
available.
Big mistakes:
Student does not use the sterile micropipette
Student does not know how to adjust the volume of the micropipette
Student tries to adjust the micropipette above its max or below (if a p200 student goes
below 20 and above 200, please watch the student if they go too far above the max for the
pipette it will fall apart)
Student does not hold the micropipette correctly
Student uses his/her hand to remove the tip from the box and then places on the
micropipette.
Student dispenses too much/too little or leaves drops in the pipette.
Student does not know how to use pipette blow out.
Small mistakes:
Student uses hand to remove tip from pipettor.
Student does not wipe pipette down with alcohol wipe
Student tries to use the yellow tips with the p1000 or vise versa
Suggested questions: As per Proctor
Scale
Students will be given the opportunity to weigh various amounts of media/salt and place
in a beaker. Various media salts will be placed next to the scale. Proctor can choose the
item to be weighed and the amount. Student will also have 2 different logs to look at.
Big mistakes:
Student does not check or fill out or use the proper log
Student does not wipe and clean the scale area before and after weighing the sample
Student does not tare the scale correctly
Student does not weight out the correct amount
Student does not clean up and dispose of items correctly.
Student does not check the expiration date or lot or item
Student does a put back (where media or salt taken out are placed back in the container)
Small mistakes:
Student makes date/other typo on the log
Suggested questions:
As per proctor
What is the meaning/definition of zeroing or tearing the scale.
When was the last time the scale was calibrated
Autodisper
Students will be asked to dispense out a certain volume 5x into test tubes. Student
should start with a dispenser that is filled with air. Machine should be off
Big Mistakes
Does not know how to calibrate
When student measures out volume it is off by more than 10%
Student does not measure out 5 aliquots.
Student does not use log
Small mistakes
Student volume is off by more than 5% but less than 10% (ex measure out 10 and
student gets 10.6)
Student makes a writing mistake on the log
