The study presented is not well designed.
General comment:

Results:
Both cancer and sarcoma are malignant tumors, but they have different
characteristics, like metastatic style, sensitivity to radio and chemotherapy. The
study tested the cytotoxity of Eugenitin on a murine sarcoma cells, but the
introduction only talk about cancer, does not mention anything about Sarcoma.
Need to write in more details for methods,
1. Cell density and Eugenitin concentration should be optimized first.
2. Either the cell culture methods or the Eugenitin concentration used in the
study may not be suitable. Since all concentrations of Eugenitin had a
similar level of effect. How were the cells counted, is it a colony assay?
Colony assay and lower Eugenitin concentration are suggested.
3. Since Eugenitin is dissolved in EtOH / PBS, pH 7.4, the control should use
same volume of EtOH / PBS, pH 7.4, instead of medium. Therefore, using
medium as control makes this manuscript very weak.
4. The time course for cell seeding, Eugenitin treatment and radiation
treatment should be described.
5. What did “under high humidity” mean? How high? ,
Again, pure extracts were dissolved and serially diluted in a 2:1 mixture of
ethanol and phosphate buffered saline (EtOH / PBS, pH 7.4). These solutions
were added as aliquots of 0.01 ml to 0.99 ml of cell culture to achieve the final
concentration of Eugenitin (0.0001 uM). The control group received 0.01 mL
growth medium and no irradiation.
1. The above sentences should be put in the methods part.
2. For the result of the second experiment, at each radiation dose should have
a control without combination of Eugenitin treatment.

For Figure 2 and Figure 3, it is better to put the control column at left side,
before the lowest concentration (uM) of Eugenitin and lowest radiation dose.

1. The methods used in this study are not suitable. Colony assay and other methods for apoptosis are
suggested.
2. Cell density and Eugenitin concentration should be optimized before real experiment.
3. Proper controls are needed. It is hard to analyze the data without proper controls
