Safe Work Procedure Title and basic
description
Title: Colorimetric detection of signal of DIG labelled probe.
Description: Detection of signal of Hybridization of DIG labelled probe to cellular mRNA using commercial
Roche kit.

Describe the activity or process
- Leave slides in Coplin Jars after stringency wash.
- Wash slides (or membrane) in 1xWashing buffer.
- Incubate slides (membrane) in 1xBlocking buffer RT 1hr containing Anti-DIG-Ab, RT, 1hr.
- Incubate slides (membrane) in 1xBlocking buffer containing Anti-DIG-Ab, RT, 1hr
- Wash slides (membrane) with Washing buffer RT, 2x15min
- Equilibrate in 1XDetection buffer.
- Develop in NBT/BCIP substrate 1.5hr to ON in the dark.
List all resources required including plant,
chemicals, personal protective clothing and
equipment, etc
Lab gown, closed shoes, gloves;
RNase free reagents; tips, pipettes and glass equipment’
Coplin jars.
Plastic “Blue lid” 50ml tubes for dilutions.

List potential hazards and risk controls
including specific precautions required
 DIG kit reagents can cause irritation to eyes and skin by contact or inhalation. Wear gloves and
gown and safety glasses when handling the kit. Must read MSDS FOR KITS before the start of the
experiment.
 Use Eye wash and shower station if contact occurs.
 NBT/BCIP substrate is light sensitive must be kept in the dark during developing step.
List emergency shutdown instructions
In the event of inhalation or ingestion the first aid officer should be consulted immediately.

List clean up and waste disposal requirements
All tips and tubes are disposed into biological waste bins.
All solutions are safe to pour down the sink. Flush with water.
