E. coli Gene Expression with Various Sugars By: Jack, Allie, Kierstin, Antoun Background ● ● ● Lactose, Sucrose, Maltose are all disaccharides Lactase breaks apart Lactose Galactose and glucose are monosaccharides Question: Do different sugars induce or repress Lactase expression? Hypothesis: We think that the disaccharides will upregulate lactose because they can fit into the active site of lactose so it can be broken down. Experimental Design Negative Control: Water instead of lactose Positive Control: Lactose Prediction: We predicted that the absorbance of the samples to which lactose, sucrose, and maltose were added would be the highest because lactase would be upregulated the most in these samples.
