SABOURAUD DEXTROSE AGAR + CYCLOHEXIMIDE CAT Nº: 1088 For the selective cultivation of yeasts and molds FORMULA IN g/l Dextrose 40.00 Cycloheximide 0.40 Peptone Mixture 10.00 Bacteriological Agar 15.00 Final pH 5.6 ± 0.2 at 25ºC PREPARATION Suspend 65.4 grams of the medium in one liter of distilled water. Mix well and dissolve by heating with frequent agitation. Boil for one minute until complete dissolution. Distribute and sterilize in autoclave at 118-121ºC for 15 minutes. AVOID OVEARHEATING as it facilitates the hydrolysis of the components and the medium remains soft. The prepared medium should be stored at 8-15°C. The color is amber, slightly opalescent. Caution: This medium is toxic if swallowed, inhaled or comes into contact with skin. Wear gloves and eye/face protection. The dehydrated medium should be homogeneous, free-flowing and beige in color. If there are any physical changes, discard the medium USES SABOURAUD DEXTROSE AGAR + CYCLOHEXIMIDE is a selective medium that can be used for cultivating yeasts, molds and aciduric microorganisms. It is also used for cultivating pathogenic fungi, particularly those associated with skin infections. This medium is at the same time used for determining the microbial and fungal content of cosmetics and for the mycological evaluation of food. The high dextrose concentration and acidic pH make this medium selective for fungi. This medium is a modification of the Dextrose Agar described by Sabouraud, with the addition of Chloramphenicol and Cycloheximide. Cycloheximide is an antibiotic which inhibits saprophytic fungi but allows for the growth of pathogenic fungi: Cryptococcus neoformans, Aspergillus fumigatus and some species of Candida (albicans, krusei). Inoculate sample and incubate at 30°C during 3 days and up yo 7 days if necessary.. MICROBIOLOGICAL TEST The following results were obtained in the performance of the medium from type cultures after incubation at a temperature of 30°C and observed after 3 - 7 days.
