Lesson Plan: Bacterial Identification Using Technology Objectives: The students will be able to: distinguish between eukaryote and prokaryote DNA explain how to create more of a DNA sample so analysis of the DNA can take place using PCR identify a specific bacterial pathogen which can cause food poisoning based upon identification of a specific sequence of DNA Activities: Day 1: Read through the case to determine what could have caused the illness o Complete virtual lab on PCR from http://learn.genetics.utah.edu/content/labs/pcr/ Set up the PCR tubes using proper techniques and protocol so that it can be amplified Day 2: o Complete virtual lab on electrophoresis using the link http://learn.genetics.utah.edu/content/labs/gel/ Take sample and transfer contents to gel and run Stain the gel Day 3: Analyze the gel to determine if prokaryotes are present Use nucleotide sequences to determine specific prokaryotes Research the 6 bacteria to determine which is pathogenic and could have caused the illness. Homework: Work on the questions in packet Complete justification for selecting the one bacteria.
