Two research efforts are ongoing in my laboratory. The goal of the first project is to produce a mobile radiation biodosimetry system for field deployment. This is being accomplished by developing and incorporating technologies in biology as well as engineering. The system will incorporate DNA probes which replicate the mRNA products of specified radiosensitive (target) genes amplified from individual human blood cells via a molecular reaction known as rolling circle amplification (RCA). The technology will be incorporated in a measuring device (optimally hand held) which uses fluorescence to quantify the mRNA products of the specified target genes. The degree of fluorescence produced by the RCA reaction will be related to the amount of ionizing radiation absorbed by the cells. The goal of the second project is to identify and characterize genetic changes associated with tumor progression and metastasis. Many types of tumors are comprised of subpopulations of cells that differ as a result of clonal expansion and concomitant accumulation of genetic changes including structural and numerical chromosome aberrations and point mutations. This tumor cell “evolution” creates a diversity of population sub-types with different potentials for invasion of nearby tissues or metastasis to distant sites. While treatment with chemotherapy or radiation therapy shrinks tumor mass by cell killing, some cells survive and many of these have accumulated additional mutations as a result of treatment. The surviving cells may have greater resistance to therapy and exhibit an associated growth advantage with adverse consequences to the patient. Understanding and characterizing these changes is critical to our understanding of tumor cell biology and is expected to be foundational to improving cancer therapies.
