PURPOSE
This procedure provides instructions for ensuring that the plasma used for
coagulation testing is sufficiently platelet poor. This procedure is to be done
quarterly.
EQUIPMENT
Abbott Sapphire or CellDyn 3200
5-10 blue top vacutainers (either 3.2% or 3.8% sodium citrate with a
proper 9:1 ratio of blood to anticoagulant.)
NOTE: Do not use hemolyzed specimens.

PROCEDURE
NOTE: TESTING FOR PLATELET-POOR PLASMA IS TO BE DONE BEFORE
TESTING FOR COAGULATION.

Action
Spin a blue top vacutainer in a centrifuge that has been programmed for
blue top tubes (5000 rpms for 7 minutes).
Using Platelet-Poor Plasma Check Log, record date, centrifuge,
accession number of specimen, RPM’s, length of spin time and your
initials.
Remove cap.
Refer to the Hematology SOP for performing patient tests on the
Hematology instrument to run in open mode. Run specimen on
analyzer.
Record platelet answer on Platelet-Poor Plasma Check Log.

Multiply the platelet answer by 1.1 to correct for the
blood:anticoagulation ratio and record answer on Platelet-Poor Plasma
Check Log.
If all of the platelet counts (post calculation) are <10,000, document that
results are acceptable.
Using form maintenance log, document the date, your initials and the
next due date.
If any result is >10,000, either another centrifuge needs to be tested for
platelet poor plasma or a variance must be filled out to continue to use
the centrifuge for blue tops. Contact Bio-Med to check the RPM’s.
