A student used the firefly luciferase as a reporter gene to demonstrate the promoter function of X gene. She
isolated the X gene promoter with 982 bp in front of the start site of transcription (+1). To define the
sequence involved in the regulation of X gene, she made a series deletions containing various lengths of the
5’ regulatory region and transfected into mammalian cells. The reporter gene activity in the absence (-) and
presence (+) of metal ion were assay and the results were showed in above figure. Read carefully and

(1). How to map the start site of transcription (+1) of the X gene?
(2). Assuming that there is a single metal responsive element (MRE) in the X gene, which fragment should
involved? Explain why?
(3). If you get the MRE sequence, how would you confirm your data by other experiments?
