Oxygen uptake rate, critical oxygen concentration Marchut-Mikołajczyk O., PhD Author/User Keywords: oxygen, uptake rate equipment required: CTN-920S Elsen oxygen sensor coupled with CO-551 Elmetron microcomputer oxygen interface execution time: 30 minutes Description: The total oxygen uptake by microbial population was determined by dynamic method ( Taguchi and Humphrey, 1966) through interrupting aeration and monitoring the decrease in oxygen concentration in the culture medium. The rate of oxygen consumption by growing bacterial cells was monitored using the CTN-920S Elsen oxygen sensor coupled with CO-551 Elmetron microcomputer oxygen interface. The maximum solubility of oxygen in culture medium was measured by determining the steady-state oxygen concentration in the not inoculated flasks. Recording of changes in dissolved oxygen concentration (dC/dt) during shortintervals of aeration (up to 10 min) resulted in the following function:dCdt¼rCx,where C is the oxygen concentration in the liquid phase (mgO2l1);t is the time (min);Cx is the cell concentration in the liquid phase (mgl1);r is the specific cell respiration rate (min1) Taguchi, H., Humphrey, A.E., 1966.
