TEACHING ACTIVITIES:

Philosophy of Teaching:
Briefly describe your philosophy of teaching and how you apply it to different types of learners, e.g. medical
students, graduate students, residents, fellows.
My teaching interests include translational biomedical and animal science and integrative medicine.
Reproductive cancers, infertility and environmental health case discussions and thought experiments are used
commonly in my graduate lectures. I use problem based learning throughout medical and graduate education
and for undergraduates where appropriate. Additionally, I enjoy teaching scientific history, comparative
human and veterinary medicine and physiology and the theory and practice of biomedical laboratory
techniques. My general teaching approach is to combine strong factual underpinnings with strategies to
engage students as active learners. I adapt this approach to match the goals of the coursework – imparting
concise basic knowledge and problem solving skills for all, emphasizing concepts for undergraduates, applied
and theoretical scientific methodology for graduate students and synthesis of pathophysiology for medical
students.

RESEARCH AND SCHOLARLY ACTIVITIES:
Opening Statement:
Provide a brief opening statement to orient the reviewers to the principal areas of scholarly activity and
provide your perspective. A description of your most significant contributions to the literature is often
helpful to the reviewers.
Cancers of the female reproductive system, endocrine disruption and infertility share a number of
suspected disease pathways and risk factors. My laboratory focuses upon the characterization and
antagonism of promising targets in animal models. Major research efforts include the study of
individuals at high risk for breast or ovarian cancer, the use of novel technologies to assess early
markers of risk for these conditions and characterization and alleviation of the premature loss of
female fertility with age and chemotherapy or toxicant exposure. A novel focus of our work concerns
the impact of selective estrogen receptor modulators on normal and precancerous breast and ovarian
tissue and the follicular reserve. I also supervise the laboratory evaluation of promising chemo
preventives for breast and ovarian cancer in human clinical trials through the KUMC Breast Cancer
Prevention Center. The central goal of this work is to improve reproductive health across species
through improved understanding of endocrine disruption and an emphasis on prevention. Major
research efforts in more detail include:
1. Breast and ovarian cancer prevention: Ovarian cancer prevention faces nearly insurmountable
biological and logistical obstacles due to difficulties in imaging and sampling ovarian tissue and the low
incidence of the disease. Our approach is to develop breast cancer prevention strategies focused on
risk factors in common with ovarian cancer. Towards this end, we have developed a novel animal
model of simultaneous breast and ovarian cancer progression which is used in parallel with our human
breast cancer prevention studies in an effort to prioritize breast cancer prevention strategies giving
simultaneous benefit to ovarian cancer. This work was recognized with a 5 year Komen Promise
Award. Additional work seeks to identify common targets for dual breast and ovarian cancer
prevention efforts. Our goal for this work is to identify and test chemoprevention agents with
simultaneous benefit for breast and ovarian cancers.




2. Human breast cancer prevention trials: Based upon my endocrine disruption biomarker work, I was
recruited into our Breast Cancer Prevention Center at KUMC under the leadership of Dr. Carol Fabian –
perhaps the most successful clinician in human breast cancer prevention in the world. For the past 7
years I have served as the laboratory scientific director of this group, directing both exploratory
endocrine and tissue biomarker analysis of our human clinical trials (including LCM, proteomic,
epigenetic, mRNA and miRNA endpoints), performing parallel preclinical studies and serving as leader
of our large cancer prevention group with Dr. Carol Fabian. Goals for this research going forward
would be to translate this successful human approach into similar efforts in veterinary species.

3. Iatrogenic oocyte loss and endocrine damage: While testing tamoxifen in our animal model of breast
and ovarian cancer, we serendipitously discovered that SERMs can protect the ovarian follicular
reserve from multiple ovotoxicants, including cancer chemotherapy drugs. We are incredibly excited
about this research since tamoxifen has already been used safely in front line cancer treatment, is
relatively inexpensive, non-invasive and avoids treatment delay – unlike current cryopreservation
efforts for fertility preservation. This work has already garnered several awards and NIH pilot funding.
An NCI R21 and March of Dimes application to explore the mechanisms of follicular rescue with SERMs
is currently in review after narrowly missing funding on initial submission. We are also performing a
locally funded teratogenesis study to determine the impact on embryo quality. Future goals for this
work are to better elucidate the therapeutic mechanism of tamoxifen during ovarian protection, to
engage human clinicians and move into human phase I study as appropriate and to apply these insights
into veterinary species. *Most recently this work was featured editorially in the journal Endocrinology
when a European group published their findings of protection from radiation-mediated infertility using
SERMs citing our studies as their incitement.





4. Ovarian physiology and endocrine disruption: This work began during my postdoctoral study with
Dr. Paul Terranova and initially focused on ovulation and its disruption by ligands of the aryl
hydrocarbon receptor in the environment. Major contributions in this arena from our group
include documentation of premature menopause from chronic, environmentally-relevant
exposures to aryl hydrocarbon receptor ligands and elucidation of major mechanisms of this
toxicity. This work garnered repeated NIEHS support and was featured at the 2010 SSR meeting in
Milwaukee. Our fertility preservation work is an offshoot of this research as well. Additionally, an
emerging topic, which we are investigating in collaboration with Dr. Margaret Petroff, is the role of
Autoimmune Regulator (AIRE) in immune-mediated premature reproductive failure. We are
resubmitting an RO1 as Co-PIs (initial score 30th percentile). This endocrine disruption work has
already been integrated into veterinary species with several grants and publications in
collaboration with European veterinary faculty.
