These techniques also include tissue special stains, frozen sections and blood smears; and differential whole fetal staining (Alizarin Red and Alcian Blue) and clearing; Morphological dissection techniques such as the Wilson freehand technique for embryonic structural evaluation, micro dissection of rodent fetuses, microsurgical removal of embryonic limb bud tissue in mice; Quantitative morphological evaluation of gross specimens (culture limbs) and microscopic tissues (e.g., thyroid, lung and bone. Semi-quantitative scoring and analysis has also been used; Reproduction evaluation techniques in rodents and lagomorphs such as synchronized breeding, determination of copulation (plug), evaluation of implantation sites, viability assessments, evaluation of post natal development markers, e.g., ear opening, hair etc; Tissue and organ cultures including embryonic limbs, fibroblast and hepatocellular cultures; Microbiology methods including general aerobic and anaerobic culture, fungal culture and special media culture for pathogens (e.g., salmonella). Quantitative assessment of colony forming units (e.g., coliform counts), antibiotic sensitivity, and enzyme analysis of cultured organisms for identification; Genetic techniques include karyotyping and chromosomal banding, genetic toxicity assessment using histidine reverse mutation (Salmonella typhimurium) and sister chromatid exchange in Chinese hamster ovary cells. Some limited experience in pedigree probabilistic assessment; Cancer evaluation including evaluation of apoptosis, dysplasia, anaplasia and neoplasia. Design and execution of rodent bioassays for carcinogenicity determination; Biochemistry including DNA, RNA and protein quantification, separation with Sefadex gels, scintillation counting of radiolabelled material (tritium and Pb).
