The Plan
-Take ADCP across mouth along with CTDP measurements
-Take CTD measurements up estuary to give idea of water column structure
- Take water samples down the estuary and do samples at every other point + take trawls at
top, bottom and middle of estuary and keep ADCP running down the estuary
- Taking ADCP at middle + head horizontally also
-1 depth profile at centre channel of estuary at top, middle + bottom
-ADCP across the mouth from time 09:31 BST
Start – 50 008 562N 005 01 068W
End – 50 08 593N 005 02 429W

Lab analysis 29/7/2011
Phosphate analysis
-
Make standard solutions with three replicates at 8 concentrations including blanks
Make reagent, make 100ml due to 78 samples
Add 1ml of reagent to each sample and mix
Leave for 1 hour
Put in U1800 spectrometer
Take 2 readings and average
Record data on sheet
Zooplankton analysis
-
Conventional methodology used to count zooplankton i.e. microscope and Bogarov
chamber to analyse 500ml samples
10ml500ml x50
Water sample = Rx0.3x0
Number m^3 = A/B
Chlorophyll analysis
The chlorophyll bottles were transferred asap to s freezer after collection aboard the Bill
Conway on the 8th June; thus preserved the samples. The following morning the samples
were ready to analyse as the action of freezing allowed the chlorophyll to be released from
the filter into the acetone solution instead of having to centrifuge the sample. Conventional
methodology was then used to calculate the concentration of the chlorophyll in each of the
samples.
To calculate the chlorophyll concentration in the water bottles:
(6/50) x value given by the flurometer where 50 was the volume of the filtered solution in ml
and 6 was the volume of acetone in ml.
NB: need to calibrate the fluorometer would normally use a known concentration of pure
chlorphyll a, approximately 90% acetone.
Offshore Boat – The Callista:
Date: 01/07/2011
Time: 7:30GMT start, 3:36GMT finish (NB all BST times converted to GMT by subtracting
an hour, and all equipment aboard The Callista worked on GMT).
Weather
Sunny, warm, calm, no precipitation and low to medium cloud cover.
Tides
Neaps, with LW at 10:26GMT, and HW at 16:13GMT.
Roles/Jobs:
Chrissy Rawlins – PSO
Michelle Barnett – Scribe and dry lab
Gareth Cheel – Equipment deployment
Ben Barton – Dry lab
Guy Hebden – Wet lab
Amelia Ballisat – Equipment deployment
Edward Gow – Wet lab
Rob Waters – Equipment deployment and shouter.
Plan:
- Using Niskin bottles on a rosette water samples were collected so DO, Silicate,
Phosphate, and Phytoplankton (Lugols) could be measured.
- Using a closing zooplankton net zooplankton samples were collected
- Using an ADCP water flow would be measured.
- Using a CTD with fluorometer and transmissometer, temperature, depth, salinity,
turbidity, and fluorescence by phytoplankton would be measured.
- Using a Secchi disk turbidity would be measured.
On the basis that a single profile using all of the above techniques to sample the water
column, would take ~45mins to complete, that Callista would travel ~10knots, and the first
station at Black Rock would take ~1hr 30mins to complete including travel from the Dock,
timings were estimated.
NB Time = Distance/Speed, where speed is in knots/nautical miles (nm) per hour, and
distance is in nm (1 nm = 1 degree of latitude).

Arrive at Black Rock (station 1) → 7:45 GMT
Depart from Black Rock → 9:00 GMT

Arrive at station 5 (around 50o55N, 005o12W – off Lizard Point) → 13:15 GMT
Leave station 5 → 14:00 GMT
Back to Falmouth Dock → 15:30 GMT
The Aim of the survey was to find fronts offshore of Falmouth, therefore as a general rule of
thumb would have to travel ~4knots, and ultimately to answer the following question:
' How do vertical mixing processes in the waters off Falmouth affect, directly and indirectly,
the structure and functional properties of plankton communities?'
