Inventory used:
- Bubble tube
- pump
- 3 clear tubes
- 1 petri dish
- soap
- water
- cassette
- 1 small screwdriver
Set up for Bubble Flow Calibration:
Cassette has an inlet connected to the bubble tube and outlet side to the pump where it reads
inlet. Put Plug pump to the wall. Put petri dish underneath the bubble tube once you have the
soap and water on them (only touch the surface of the water). Calibrate by using a screwdriver
to move the flow to 1L/ minute. Use a cap on the top of the bubble tube to cover the airflow
from the top of the bubble tube.
1L/minute:
First Run
1000mL to 0mL: 75.00 seconds
Second Run
1000mL to 0mL: 74.80 seconds
Third Run
1000mL to 0mL: 73.14 seconds
Calculate Flow Rate
First Run (1L/min): Volume/Time: 1L / 1.25 (75/60seconds) = 0.80
Second Run (1L/min): Volume/Time: 1L/ 1.25 (74.80/60 seconds) = 0.80
Third Run (1L/min): Volume/Time: 1L/ 1.22 (73.14/ 60 seconds) = 0.82
Average Time: 0.81L/minute
1.5L/minute:
First Run
1500mL to 0mL: 74.42 seconds
Second Run
1500mL to 0mL: 74.11 seconds
Third Run
1500mL to 0mL: 74.48 seconds
Calculate Flow Rate:
First Run (1.5L/min): Volume/Time: 1.5L/ 1.24 (74.42/60 seconds) = 1.21
Second Run (1.5L/min): Volume/Time: 1.5L/1.24 (74.11/60 seconds) = 1.22
Third Run (1.5L/min): Volume/Time: 1.5L/1.24 (74.48/60 seconds) = 1.22
Average Time: 1.22 L/ minute
2L/minute:
First Run:
2000mL to 0mL line time: 63 seconds
Second Run:
2000mL to 0mL: 74.96 seconds

Calculate Flow Rate:
First Run (2L/min): Volume/Time: 2L/ 1.05 (63 seconds/60 seconds) = 1.90

Third Run (2L/min): Volume/Time: 2L/ 1.23 (73.56/60 seconds) = 1.63
Average: (1.9 + 1.6 + 1.63/ 3) = 1.71 L/ minute
SOP #2: Defender 510 Pump
Put the tube to the suction section of the defender pump and the other end to the cassette
(inlet); put another tube on the outlet end of the sample pump and the other end to the
cassette end of the sampler pump.
Place the pump setting to continuous
