Heartland Community College Fall 2011 - Biology 191 – Introductory Microbiology Lab Lab Sections L2 – TR 9:00 – 10:50 AM L5 – TR 1:00 – 2:50 PM Instructor Information: Jon Hoekstra Office: 2414 ICB HCC Phone – 309-268-8648 E-mail – jon.hoekstra@heartland.edu Office hours – MTWR 11:00 AM – 12:00 PM; MW 4:00 – 4:50 PM Laboratory Manual Information Students may obtain lab exercise instructions, pre-lab assignments, and final lab reports by printing instructions from the course website at this address: http://employee.heartland.edu/SROSE01/BIOL191/Lab/lab.index.htm Notice of Cancelled Class Sessions Cancelled class sessions for all HCC classes will be listed under Cancelled Class Meetings in the A-Z Index and under Academic Information in the Current Students page on the HCC Web site. Go to http://www.heartland.edu/classCancellations/ to learn what classes have been cancelled for that day and the upcoming week. Be sure to check the last column, which might contain a message from the instructor. Pre-lab Reports, Lab Reports, and Exams Excluding the Microscopy, Epidemiology, CD-ROM labs, exercises contain two assignments, the PREREPORT and the LAB REPORT. The Pre-Report should be completed before the day the exercise BEGINS. It will be collected AT THE BEGINNING of the lab period before the pre-lab instruction. The purpose of the pre-report is to encourage students to read the lab before coming to class. It will not be accepted late. The Final Lab Report should be completed and turned in immediately after the lab is completed. Because lab work is “hands-on”, your presence in class is necessary. Therefore, if you are absent, the pre-report and/or lab report due that day will not be accepted. There are no make-up labs. The laboratory portion of the course comprises 25% of the total BIOL 191 grade. However, a grade of 70% or higher in the lab portion is required in order to pass the course. Failure to achieve 70% or more in the lab portion will result in a grade of “F” for the entire course regardless of the grade in the lecture portion. Points for the laboratory portion of the course: Assessment Microscopy Lab Report Pre-reports (10 @ 5pt. Each) Lab reports (11 @ 5pt. Each) CD-ROM labs (2 @ 5pt. Microbiology Lab Schedule Fall 2011 Date Week 1 8/23 8/25 Exercises PL = prelab due; LR = report due Introduction to the lab Video: Bacteria Microscopy LR Day 1 - Widespread Distribution PL Objectives Lab schedule, safety regulations, laboratory equipment and student evaluation are discussed. Principles of microscopy are introduced. Students learn to utilize the compound microscope and immersion oil techniques. Video: The Tree of Life Students examine the diversity and ubiquitous nature of bacteria. Aseptic technique is introduced. 9/1 Day 2 - Widespread Distribution of Microorganisms LR Students learn simple staining techniques and observe bacterial cells using the microscope. Week 3 9/6 LABOR DAY – NO LAB (Tuesday LABS are “off” to maintain weekly schedule) 9/8 Day 1 - Pure Cultures & Aseptic Tech. PL Week 2 8/30 Culturing procedures are performed utilizing a mixed culture. QUIZ 1 Week 4 9/13 Day 2 - Pure Cultures & Aseptic Technique CD-ROM: Biochemistry: The Chemistry of Living Things WS Day 3 - Pure Cultures & Aseptic LR 9/15 Week 5 9/20 Video: E. coli: Confessions of a Germ Day 1 - Sampling Food, Water, Milk PL Day 1 - Intestinal Pathogens PL Day 2 - Sampling Food, Water and Milk LR Students observe colonies from mixed samples and attempt to isolate pure cultures. Students perform interactive tutorials. Students observe pure cultures and view them under the microscope. Sampling procedures are performed. Students attempt to isolate intestinal pathogens on selective media. Colony counts and presumptive determination of contaminating microorganisms are done. 9/22 Day 2 - Intestinal Pathogens continued Fermentation tests are performed on selected intestinal pathogens Date Exercises PL = prelab due; LR = report due Objectives QUIZ 2 Week 6 9/27 9/29 Week 7 10/4 Day 3 - Intestinal Pathogens continued Final inoculations will be performed on differential media Day 1 - Control of Microbial Growth: Alcohol & Hand Scrubbing PL Students begin testing the effectiveness of alcohol swabs and hand scrubbing Day 4 - Intestinal Pathogens LR Final evaluation of isolated intestinal pathogens Day 2 - Control of Microbial Growth: Alcohol & Hand Scrubbing LR Day 1 - Control of Microbial Growth: Evaluation of Antimicrobics PL Video: Super Staph: Invincible Bacteria? Day 2 - Evaluation of Antimicrobics LR 10/6 Week 8 10/11 Day 1 - Isolation of an Antibiotic Producer from Soil PL Day 2 Isolation of an Antibiotic Producer from Soil continued Students evaluate the effects of alcohol and hand scrubbing Students begin tests to determine the effectiveness of specific disinfectants, antiseptics, and antibiotics Students evaluate the effectiveness of the antimicrobics. Students begin primary isolation of antibiotic producing bacteria from soil Students will select antibiotic producing colonies from soil cultures Video: Creators of the Future QUIZ 3 10/13 Week 9 10/18 Day 3 - Isolation of an Antibiotic Producer from Soil LR Day 1 – Transformation PL CD-ROM: Microbes in Motion WS Students will observe soil cultures for evidence of antibiosis Students will begin experiments which will lead to the genetic transformation of E. coli Students will perform interactive tutorials Day 2 – Transformation LR Students will observe transformed cultures DNA Extraction LR Students will isolate bacterial DNA 10/20 Date Exercises PL = prelab due; LR = report due Objectives QUIZ 4 Week 10 10/25 Day 1 - Preparing Unknown Cultures: Morphological Characteristics PL Morphological characteristics of an unknown bacterial culture are examined. 10/27 Day 2 - Preparing Unknown Cultures: Morphological Characteristics LR Morphological characteristics of the unknown are examined and described. Week 11 11/1 Day 1 - Unknown Cultures: Physiological Characteristics PL Students test biochemical characteristics of their unknown. Students become familiar with problems involved in determining the identity of an unknown and learn about resource manuals. 11/3 Day 2 - Physiological Characteristics of the Unknown LR Students complete biochemical testing of their unknown. References needed to determine the unknown are utilized.
