MICROBIOLOGY TEST #1 This is just a guideline to help you study. Information from lab manual, handouts (look at my website www.jcapers-irsc.weebly.com , lecture or discussion in class, as well as lab resources online will be covered. The exam will consist of two parts: short answer (52 points) and lab practical (48 points), total of 100 points Short answer essay Week 1: Know the parts and function of microscope Define (and/or) calculate total magnification, resolving power, parfocal, numerical aperture Know how to use and care of microscope Know basic shapes of bacteria Week 2: Know the different methods to determine motility Understand true motility versus brownian movement, and water movement How does dark field microscopy work? Know how to change from bright field to phase contrast and dark field Distinguish and compare advantage/disadvantage between bright field, phase contrast, and dark field microscope Can a bacteria have more than one flagella? Of the 2 bacteria we tested, which one has true motility? What were the results of the agar method? Week 3: Know the different ways to obtain a pure culture (spread plate, streak plate, pour plate) How do you make a streak plate? Know how to make a smear on a slide from specimen growing on agar (solid) and a broth (liquid) Understand why stains work. Do basic stains work better? Why? Know different between simple stain and differential stain Week 4: Know the procedures for doing the gram stain and acid fast stain, know what happens at each step (don’t worry about times) Understand the concept of how both the gram stain and acid fast stain work Know the differences in morphology of cell wall of gram positive and gram negative bacteria Know the differences in morphology of cell wall of acid fast positive and acid fast negative bacteria
