872 Main Street Buffalo, NY 14202 (800)274-5487 fax (716)882-0959 PRODUCT DESCRIPTION SIV Mac 251 PURIFIED VIRAL LYSATE Catalog # 0810011 Size 1.0 mg The SIVMac 251 (Simian Immunodeficiency Virus) is propagated in Hut78, a human peripheral blood T-cell line. The virus is purified using density gradient ultracentrifugation, disrupted in the presence of detergent and characterized at the biochemical and immunologic levels to assure product consistency and performance. PRODUCT COMPOSITION 3 Purified SIV Mac 251 (density 1.16 g/cm ) is disrupted in 0.5% Triton X-100 nonionic detergent/0.6M KCl. Detergent is removed by 2 cycles of ether extraction and the virus is subjected to in vitro testing to ensure lack of infectivity. Each lot of purified virus is examined for purity using SDS-PAGE and for antigen composition using Western immunoblotting (WIB). Diagnostic levels of env proteins (gp160, gp120, gp41), gag proteins (p17, p24, p27, p39, p55) and pol genederived proteins (p31, p51/p66) are present in the virus. The total protein concentration is determined by Pierce BCA assay. Verification of virus inactivation is determined by Reverse Transcriptase Reduction Assay. RECOMMENDED USES Immunodetection of antibodies to SIV using solid-phase enzyme immunoassay (EIA), WIB, dot-blotting and related techniques. The material should also be of value as an immunogen and for the purification of viral proteins. STORAGE AND STABILITY o The purified virus is stable for at least one year when stored at –60 C or below. To avoid freeze-thawing, unused material should be stored in aliquots, preferably under nitrogen in order to prevent oxidation. AVAILABILITY Distributed in vials containing 1 mg of protein, shipped on dry ice. Protein concentration ranges from 0.5 to 1.5 mg/ml. Custom orders are available, including specific buffer formulations and package sizes. All products are for Research Use Only and not for in vitro Diagnostic Use This Package Insert is provided for product evaluation purposes only and is not intended to be used in place of the Package Insert shipped with the product.
