Please RSVP with number of attendees to minnmass@mac.com PAGE 1 of 4 MINNESOTA MASS SPECTROMETRY DISCUSSION GROUP SEPTEMBER 22, 2008 ABSTRACT The characterization of large biomolecules such as proteins by mass spectrometry was greatly facilitated by the development of desorption/ionization methods, such as matrix-assisted laser desorption/ionization (MALDI) and electrospray ionization (ESI). One of the hallmark features of ESI is the multiple charging phenomenon that allows large molecules to be measured using analyzers of moderate mass-to-charge ratio (m/z). Multiple charging allows also efficient dissociation of intact proteins (i.e., top-down mass spectrometry) for sequencing, identification, and detection of posttranslational modifications. New ambient methods for multiple charging of proteins desorbed from solids potentially offers more efficient means for protein identification from tissues. Increasing charging beyond that generated by normal ESI processes can be exploited for the analysis of protein-ligand and protein-protein complexes. Reducing multiple charging can be advantageous for the detection of very large proteins up to the MegaDalton size, especially when coupled to ion mobility analyzers. BIOSKETCH OF PROFESSOR JOSEPH A. LOO Dr. Joseph A. Loo is a Professor in the Department of Biological Chemistry, David Geffen School of Medicine, and in the Department of Chemistry & Biochemistry at the University of California, Los Angeles (UCLA), and he is the Faculty Director of UCLA Jonsson Comprehensive Cancer Center Mass Spectrometry and Proteomics Technology Center. He is also a member of UCLA/DOE Laboratory for Genomics and Proteomics, the UCLA Molecular Biology Institute, and the UCLA Jonsson Comprehensive Cancer Center. He is an expert in the mass spectrometry characterization of proteins and their post-translational modifications, and is the author of over 180 scientific publications. He has been on the Editorial Boards of the journals Bioconjugate Chemistry, Analytical Chemistry (A-Page), Journal of the American Society for Mass Spectrometry (JASMS), and Chemical & Engineering News and currently he serves on the Editorial Advisory Board for Rapid Communications in Mass Spectrometry and as an Associate Editor for JASMS. In 2000-2002, he served on the Board of Directors for the American Society for Mass Spectrometry. His research interests include the development of bioanalytical methods for the structural characterization of proteins and their application for proteomics and disease biomarkers. Currently, his laboratory is developing technologies to profile proteins in human salivary fluids for their application for cancer biomarker discovery. He is also interested in the development and application of mass spectrometry for the study of noncovalently-bound macromolecular assemblies and their interactions with other binding partners and ligands. At UCLA, Dr. Loo teaches a course for undergraduate students on “Chemical Instrumentation” and a graduate level course on “Proteomics and Mass Spectrometry.” Before he joined UCLA in 2001, he was Group Leader of the Biological Mass Spectrometry and Proteomics Teams at Parke-Davis Pharmaceutical (currently Pfizer Global Research), Ann Arbor, MI. He worked at Warner-Lambert/Parke-Davis/Pfizer for nearly 10 years before moving to UCLA. He received his BS degree in Chemistry from Clarkson University (Potsdam, NY), and his Ph.D. degree in analytical chemistry from Cornell University in 1987 with Professor Fred W. McLafferty, working on the development of Fourier transform mass spectrometry for bioanalytical applications. He carried out postdoctoral research at Pacific Northwest National Laboratory, Richland, WA with Dr. Richard D. Smith on the development of electrospray ionization mass spectrometry and capillary electrophoresis for protein characterization.
