Specificity and Comments
Human CD10, also known as common acute lymphoblastic leukemia
(CALLA), has been shown to react with TdT+ lymphoblastic
leukemia, follicular germinal cell lymphoma, Burkitt’s lymphoma,
and chronic myelocytic leukemia. CD10 also marks normal early
lymphoid progenitor cells, immature B-cells in adult bone marrow,
and germinal cells in normal tonsil and/or normal lymphoid tissue. It
is also expressed in some non-lymphoid tissues such as fibroblasts,
breast myoepithelium, and brush border of kidney. Recently, CD10
has been used in a panel for mantle cell lymphoma with cyclin D1(+),
CD43 (+), CD5 (+), IgM (+), CD23 () and CD10 ().
ReVeal maybe substituted, but will require longer incubation times
with the CD10 primary antibody. Allow the solution to cool for 20
minutes.
Alternatively, steam tissue sections for 45-60 minutes, or use a
pressure cooker for 2-3 minutes
(BIOCARE’S Decloaking
Chamber). Cool for 10 minutes.
Protein Serum Block (Optional)
Incubate for 5 minutes at room temperature (RT).

Dilute 1:25-50. Incubate for 30-60 minutes

Incubate for 10 minutes at RT with BIOCARE’S 4plus Detection
System.

Enzyme Labeled Streptavidin
Prokaryotic recombinant fusion protein corresponding to the external
of the CD10 glycoprotein.
Incubate for 10 minutes at RT with BIOCARE’S 4plus Detection
System.
Cellular Location: Membrane
Chromogen
Control Tissue:
Incubate for 5 minutes at RT with BIOCARE’S
CHROMOGENCARE.

Technical Note
Known Applications:
CD10 is a cell surface marker that may show low expression in
certain tumors and/or tissues. Overstaining with hematoxylin can
mask staining, thus reducing overall contrast and making
interpretation difficult. It is recommended that a light hematoxylin
stain be applied (15 to 30 seconds with Tacha’s Hematoxylin). If
using high pH retrieval solutions, some tissues may require an avidinbiotin block.

Protocol Notes
Protocol Recommendations:
The optimum antibody dilution and protocols for a specific
application can vary due to many factors. These include, but are not
limited to: fixation, incubation times, tissue section thickness and
detection kit used. The data sheet’s recommendations and protocols
are based on exclusive use of BIOCARE products. Due to the
superior sensitivity of these unique reagents, the recommended
incubation times and titers listed are not applicable to other detection
systems, as results may vary. Ultimately, it is the responsibility of
the investigator to determine optimal conditions.
Endogenous Peroxidase Block
If using an HRP system, block for 5 minutes with BIOCARE’S
PEROXIDAZED 1.
