Project – Monoclonal Antibody Production and Hybridoma Technology
***Details - Injected Listeria monocytogenes antigen into Balb/C mice for antibody production, collected immune
splenocytes, fused with tumor cells. Fused cells were selected, screened, cloned. Learnt cell culture and aseptic
techniques in detail. Performed a number of ELISA assays during the screening process. Primed the Balb/C mice with
pristane for Ascites production. Injected hybridoma cells. Tapped cells from mouse, purified the immunoglobulins by
column chromatography, characterized the antibodies. Purity was determined by SDS-PAGE. Densitometry analysis was
carried out using Kodak Molecular analyst software. Specificity of monoclonal antibody was determined using Immunoblot
analysis. Performed monoclonal antibody isotype determination using Flow cytometry. Recorded data systematically in
lab notebook.

***Part I - Model system used – maize plant light regulated genes. Learnt to design PCR primers, Isolated DNA from light
grown and dark grown maize shoots, Quantification of Maize DNA by gel analysis, Optimized the maize gene PCR,
Purified PCR products, Biotinylated the PCR products, Isolated total RNA from maize seedlings, gel analysis of RNA,
performed Northern blot analysis of maize gene expression, performed Hybridization and Chemiluminescent detection of
Northern blot, PCR Trouble shooting PCR and Northern blotting.
***Part II - Extraction and determination of Maize seedling protein, Isolelectric focusing, SDS-PAGE ***D electrophoresis.
