Klug, NEW Activity 3.4 Analysis of Human Pedigrees
* In its revised format include parts 3.4.1 through 3.4.5. These parts should be separated
from the original tutorial then combined to form this new tutorial, Activity 3.4. It will
have it's own Introduction, Summary, and Post-quiz.
* Remove the screen that shows on selecting “module quiz” asking if the user wishes to
take the quiz.
New organization:
Activity 3.4, Analysis of Human Pedigrees
Introduction (New 3.4.1.1)
- This will need to be a Flash animation.
- Delete the stops between successive pages of text.
- Use the same image as in old 3.4.1.1 of the pedigree chart and human eyes
photo.
- - Scroll the text from old 3.4.1.1, page 1, “Pedigree analysis is used to
evaluate….” from the bottom of the text window to the top. When the top
line reaches the top, freeze the scrolling, pause 5 seconds, and dissolve a
scrolling of the next page of text, 3.4.1.2.
- Scroll the second page of text saying “The illustration at left is of a
hypothetical pedigree….” from the bottom of the text window to the top,
stopping when the top line reaches the top. Pause for 4 seconds and dissolve
to the scrolling of the third page of text, 3.4.1.3.
- Scroll the third page of text saying “The proband (male propositus, female
proposita) is….” from the bottom of the text window to the top, stopping
when the top line reaches the top. Pause for 4 seconds and dissolve to the
scrolling of the last page of text, 3.4.1.4.
- Scroll the fourth page of text (3.4.1.4) saying “In this example, the proband is
heterozygous…” from the bottom of the text window to the top, stopping
when the top line reaches the top.
- This is the end of the segment.
- Set scroll flags at the start of 3.4.1.2 and call it “Pedigree chart”, a the start of
3.4.1.3 called “Proband defined”, and the start of 3.4.1.4 called “Example”.
Exercise(New 3.4. 2) Single Character Pedigree Acitivty
- Begin directly with the activity image showing drop-down windows next
to the pedigree chart entitled “Pedigree for a Single Character” as in old
3.4.2.2. Delete the old beginning image of 3.4.2.1 that showed the chart
without the drop-down windows.
- Change the Score button into a SUBMIT button.
- Delete the small instructions by the SCORE button that say “Make a
choice for each menus…..”
- Keep the same interactivity and popups that occur in the extant exercise.
- Delete the Exercise Instructions box that appears and move the text of this
box (“Determine the possible genotypes….” into the Interaction
Instructions box in our new format. Change these instructions slightly so
that the third line instead says “When you are finished, click the Submit
button to see how well you did.” [i.e. change “Score” to “Submit”]
- At the very end of the instructions in the Interaction Instructions box, add
the following line:”Click on the CONTINUE button to see the chart with
all the correctly assigned genotypes”.
- Add a CONTINUE button to the popup Interaction Instructions which
dissolves to the second image, 3.4.2.2.
- In 3.4.2.2, bring up the Pedigree chart image that shows all the answers
(i.e. “aa, AA, Aa”) over each box or circle.
- This is the end of the segment.
Animation (Parts 3.4.3) X-linkage in Humans
- In 3.4.3.1, (titled X linkage in humans), use the same extant graphic of the
pedigree chart. Scroll the two paragraphs from the first page of text
“Inheritance of some traits…” from the bottom of the text window to the
top, freezing the scroll when the top line reaches the top. At this point,
pause 3 seconds and dissolve to the scrolling of the next page of text,
3.4.3.2 .
- Scroll the next page of text (“Two important human conditions are…”)
from the bottom of the text window to the top until the top line reaches the
top. This is 3.2.4.2. Freeze the text at this point, pause 3 seconds, and
dissolve to the scrolling of the next page of text, 3.4.3.3 .
- Scroll the next page of text (“The panel on the left depicts…”) from the
bottom of the text window to the top until the top line reaches the top.
This is 3.2.4.3.
- This is the end of the segment. Delete the last page of text associated with
this segment.
- Separate the s interactive activity in the old X-linkage in Humans into a
new segment, 3.4.4 (below)
-Set scroll flags “X linked examples” at 3.4.3.2 and “Chart” at 3.4.3.3.
Exercise (Part 3.4.4): X-linkage in Humans
- Take the interactive part of old 3.4.3.5 and make it self-contained.
- This is the chart pedigree diagram where the user must pull down several
drop down windows and make selections, then press “Score”.
- Place the following instructions into the Interaction Instructions pop up:
“Draw down the list of possible genotypes for each individual by clicking
on the black triangle arrows. Assign each individual of the genotypes in
the box by clicking on that genotype. Then click the Submit button to
check your answers.” .
- Delete the Exercise Instructions box that comes up.
- Delete the text in the graphics window under the chart saying “Make a
choice for each menus and then….”
- Change the Score button to a Submit button.
- In the textbox, have scrolling text moving upwards beginning at the
bottom. Delete the extant text and replace with:
“A partial pedigree of hemophilia is presented in this activity
which indicates how this trait descended from Queen Victoria in the British
royal family. Historical research has concluded that X-linked hemophilia was
not present in any of Queen Victoria’s ancestral lines, but careful
consideration of the pedigree leaves little doubt that she was heterozygous for
the trait. By convention, circles with dots in them, or circles that are half
filled and half empty, are used to represent females presumsed to be
heterozygous for the trait (carriers).
Use your knowledge of X-linked traits to assign the most probable
genotypes for each individual in the adjacent pedigree. It may not be possible
to assign a genotype uniquely in all instances based on the available
information’ the possible choices include ambiguity in the assignment in some
cases.”
[Stop the scrolling when the paragraph beginning “Use your knowledge of
X-linked traits…” reaches the top.]
- Use all the extant interactivity for this exercise.
- When the user has clicked on the SUBMIT button and gets the feedback
of red Xs and/or checkmarks (we’ll call this 3.4.4.2), a pop up window
says “That completes the exercise…” Change this popup box to say “That
completes the exercise. You may repeat it by clicking the Reset button.
Otherwise, click CONTINUE to see all the answers”. Add a CONTINUE
button to this window that dissolves from this activity to the next screen
(which I’ll call 3.4.4.3).
- In 3.4.4.3, bring up the new image of the completed chart as well as the
text “The most likely genotype assignments….”
- This is the end of the segment/exercise
- No scroll flags.
Summary (New 3.4.5)
-This will not be programmed as part of the Flash animation.
Post-quiz (New 3.4.6)
-This will not be programmed as part of the Flash animation.
- Delete screen asking if user wants to take the quiz.

1. In the Introduction, which is a flash animation in this tutorial, there are errors in
the second unit of scrolled text which begins “The illustration at left is of a
hypothetical….” The errors are in the first sentence. It should properly read “The
illustration at left is of a hypothetical pedigree for eye cataracts in humans, an
autosomal dominant trait.” (bold added just for editing emphasis).
[Andy: Correct the sentence as directed.]
2. Make the play screen scalable in the user’s browser and allow it to fill the whole
browser window.
[Andy: Yes, all screens should be scalable.]
3. In 3.4.2, the Pedigree for a Single Character Exercise, delete the on-screen
instructions in the lower right corner which begin “Make a choice for each menus
and then click….”
[Andy: Yes, remove this text. Also, the Interaction Instruction box should be
open at the beginning by default.]
4. Delete the CONTINUE button that appears after the user clicks on the SCORE
buttonin 3.4.2. It doesn’t do anything except delete all the interactive areas of the
screen!
[Andy: Also, remove the text box that reads “That complete this exercise. You
may repeat it by clicking the Reset button.” This box partially obscures the
results of the exercise.]
5. In 3.4.4, the X-Linkage in Humans Exercise, we need to add a blank line between
the first and second paragraphs of the scrolled text. This blank line comes just
before “Use your knowledge of X-linked traits to assign….”
[Andy: Yes, paragraphs should be separated by a blank line. Also, the Interaction
Instruction text refers to a “Submit” button but the button is titled “Score” and not
“Submit”. Please correct this. Next, remove the text box at that reads “That
complete the exercise. You may repeat it by clicking the Reset button.” This text
box obscures the results of the exercise. Finally, the Interaction Instruction box
should popup up AFTER the text narrative finishes scrolling.]
6. It just occurs to me that maybe Lightcone isn’t done yet with the first pass on this
tutorial even though it was listed as ready as of 10/5/03. I can’t progress to the
conclusion from the X-linkage in Humans Exercise. The Next button just restarts
this last exercise. Is the conclusion done and not yet hooked up to the end?
[Andy: Please advise on the completion of the Conclusion page so that it can be
reviewed.]
Andy’s Comments 10/7/03:
* There must be an Introduction and a Conclusion in HTML format. This is the page
that loads in the WPS window to introduce the tutorial. In all cases where Eric has
directed the creation of a Flash Introduction, we need to assemble an HTML
precursor.
* For the HTML Introduction, use this text: “Pedigree analysis is used to evaluate the
genetic basis of phenotypic traits in organisms where controlled experiments to
generate large numbers of offspring are not practical or ethically possible.” Remove
this same text from the first page of the Flash file. Change the first scroll-bar flag to
“Overview.”
* The Conclusion text is in the old tutorial and listed as the “Module Summary.”
Please add this unnumbered text to the Conclusion for this tutorial.
Second Pass Comments from Eric Stavney 10/29/03
1. To be consistent, we should change the title of the Conclusion page to just
“Conclusion” instead of “Analysis of Human Pedigrees Summary”.
