Background: Rotaviruses are the most common cause of dehydrating gastroenteritis in infants and children, and
also in a variety of mammals and birds. Rotavirus is a double-stranded (ds) RNA virus of the family Reoviridae and
is classified into 8 groups (A to H) based on antigenicity and nucleotide sequence identities of the VP6 gene. Group
A causes approximately 90% of infections in humans and animals, group B causes gastroenteritis primarily in adults
and is mainly found in China. Group C is found worldwide but this virus is extremely less prevalent, while group D, E
and F has not been found in humans. The virus is composed of three concentric shells that enclose 11 gene
segments. The outermost shell contains two important proteins – VP7 (G genotypes), and VP4 (P genotypes). The
antigenicity and gene sequence diversity of these proteins define the two serotypes (G-serotype and P-serotype)
and genotypes (G-type and P-type) of the virus. To date, at least 27 G and 37 P genotypes have been identified in
humans and animal species.
Description: ViroReal® Kit Rotavirus (A) is based on the amplification and detection of a part of the non-structural
protein 4 (NSP4), a transmembrane ER-specific glycoprotein, using one-step reverse transcription real-time PCR. It
allows the rapid and sensitive detection of RNA of rotavirus (genotype A) from samples purified from fecal specimen
(e.g. with the QIAamp Viral RNA Mini Kit).
PCR-platforms: ViroReal® Kit Rotavirus (A) is developed and validated for the ABI PRISM® 7500 instrument (Life
Technologies), LightCycler® 480 (Roche) and Mx3005P® QPCR System (Agilent), but is also suitable for other realtime PCR instruments.
Specificity: ViroReal® Kit Rotavirus (A) is specific for rotavirus isolates belonging to genotype A.
