The 1 kb plus DNA ladder was the standard that was referred to for the molecular weight. This standard molecular weight marker was used to generate a standard curve comparing fragment size (bp) against the distance each fragment traveled in centimeters (Figure 2). From this standard curve, the sizes of the vector sequence and cDNA insert can be determined. According to the graph, the vector sequence is approximately ----- and the cDNA insert is approximately 1000 bp (Table 1). This cDNA insert was then sent off to Ohio State for sequencing.
