AutoMACS™ Pro FAQ—1 Cell Rescue Procedure This procedure is designed to recover cells from the autoMACS™ Pro Separator if a power failure has occurred, or if an error has been detected requiring the unit to be powered down (e.g. pump overload error on autoMACS™ Classic Separator). If an operation error has been detected contact technical support 1-800-367-6227 for further assistance. 1) Turn the autoMACS™ Pro Separator off if the operation has ceased due to an error detection. If power was interrupted, turn the power button to the off position and await power restoration to the outlet. 2) Determine whether the automatic rinse function upon start up is enabled on the autoMACS™ Pro Separator. If you do not know, assume that it is. 3) Power the unit up and allow the initialization to perform. You will receive a message indicating that the unit was not shut down appropriately. Select OK. 4) If the automatic rinse function has been DISABLED, proceed to step 5. If the automatic rinse function is ENABLED, then following completion of the initialization, stop the Rinse function by touching the ABORT button on the screen. Now proceed to step 5. 5) Unscrew the waste bottle closure. Wipe the outlet port clean with 70% EtOH and place into a sterile 50 ml collection tube in a tube holder. 6) Remove the uptake needle by gently pushing out of the holder. See figure 1. Wipe clean with 70% EtOH and place into a sterile 15 or 50 ml collection tube in a tube holder. 7) To gain access to the negative elution port, rotate the wash station access panel to the left. Unscrew the wash station from the autoMACS™ Pro Separator by turning the thumb screw to the left. See figure 2. Gently pull the wash station forward and lay out of the way to the right. Hold a sterile 15 or 50 ml collection tube under the negative elution port. 8) Activate the Separation Screen and select the Wash Only function. 9) Choose QRinse. 10) Wait for all the buffer and cells to be dispensed into the three tubes. a. Waste ~35mls b. Negative ~7mls c. Positive ~10mls 11) Combine all tubes and spin down cells to reapply for cell separation either on a manual column system or on the autoMACS™ Pro Separator. 12) Return the wash station to normal position by using the small pins as guides and screw the unit to the autoMACS™ Pro unit. 13) Return the needle arm into the holder and gently snap into place (Figure 3). 14) Reattach the bottle closure to the waste bottle. 15) Run a Rinse program. 16) If the autoMACS™ Pro Separator is operating normally, repeat cell selection. Alternate Procedure: This one may be easier to perform, but I think may lead to more issues with tube stripping and contamination from the negative port. 1.
