SYBR-SAFE PROTOCOL
SYBR-SAFE is a non-toxic chemical that is used to visualize DNA under UV light in
agarose gel electrophoresis. It is a great substitute for ethidium bromide, which is an
extremely dangerous chemical. SYBR-SAFE is sensitive to light so we wrap the gel box
in aluminum foil both when preparing and running gels.
SYBR-Safe can be purchased from Invitrogen. It is stored at room temperature and comes
in a 400 ul tube (Product # S33102 for $47).
-The product is at 10,000X concentration in DMSO buffer.
-Shu-Dan Yeh (True Lab at Stony Brook University, Gmail correspondence Nov 1, 2008)
indicated that it should be diluted by 10X to 1000X concentration in 0.5X TBE buffer.
-We add 8 ul to 40 ml of TBE buffer for 1-2.5% agarose gels and get very good results.
On Sep/14/09 WAM prepared a working batch by adding 50 ul SYBR Safe to 450 ul
0.5X TBE buffer. This is our working stock.
