AMP-int-R and LEU2-int-F were designated as specific primers matching the up- or
down- stream region of PMT1 ORF or the vector sequence, and URA3-int-F,
Verify-hac1-F and Verify-hac1-R were designed from the disruption cassette (URA3)
or the up- or down- stream region of HAC1 ORF. b. PCR products amplified from
genomic DNA of pmt1hac1 strain. The 1814 bp band in lane 1 was amplified with
Verify-pmt1-F and AMP-int-R primers; the 1834 bp band in lane 2 was amplified with
LEU2-int-F and Verify-pmt1-R primers; the 691 bp band in lane 3 was amplified with
URA3-int-F and Verify-hac1-R primers; and the 1657 bp band in lane 4 was amplified
with Verify-hac1-F and Verify-hac1-R primers. M represents DNA marker.
