“Sensitive detection of intracellular sumoylation via SNAP tag-mediated translation and RNA
polymerase-based amplification”
This article aimed to detect mutant O-6-alkylguanine-DNA alkyltransferase. The mutant is incorporated
into a double-strand of DNA and transcripted via T7 RNA polymerase to amplify the RNA product. Upon
addition of RiboGreen, an RNA intercalating dye, yields fluorescent detection. The RNA translated
results have potential for detection of small modifiers to target proteins.

“Sensitive and label-free DNA methylation detection by ligation-mediated hyperbranched rolling circle
amplification”
This article explores specific detection using hyperbranched rolling circle amplification instead of PCR,
enzymes, or fluorescent labels. The target is methylated DNA, which can lead to early cancer diagnosis
at CpG sites on genomic DNA. The method detected up to 0.01% level of methylation using fluorescent
dye SYBR Green I.

“On-chip aptamer-based sandwich assay for thrombin detection employing magnetic beads and
quantum dots”
This study used thrombin as the target analyte in a unique assay based on the sandwich ELISA. Two DNA
thrombin aptamers attached to different epitopes on thrombin. The sandwich was completed by
attaching magnetic beads for capturing the analyte were attached to one aptamer, and the other was
functionalized using quantum dots. Fluorescence spectroscopy was used to monitor the assay.
