Project Objectives
Short term:
Assess diffraction of crystals made from antibody-ligand complexes and membrane proteins involved
in egg-sperm interactions. Collect diffraction data on ligand complexes of poly-ADP ribose polymerase
proteins.
Medium term:
Determine the 3D structure of autoantibodies and poly-ADP ribose polymerases in complex with their
ligands and membrane protein structures involved in human fertilization.
Long term:
Improved understanding of autoimmune diseases, human fertilization and cancer.

Project Achievements for this Visit

Tested several crystals and collected several datasets of various ligands - unfortunately only to around
3Å resolution. Refinement in progress.

Confirmed membrane protein crystals however crystals diffracted to around 10Å at best.
6377.16
Test crystals resulted in a 4.2Å resolution dataset when auto-processed by Diamond. Crystals was
reprocessed with optimal settings and fewer frames since the later part of the data suffered from
radiation damage and structure of a mouse antibody in complex with a peptide has recently been
solved to 3.1Å resolution.
