CRISPR/Cas9 RNA Injection
Description: The Core will inject purified RNAs into the pronucleus and cytoplasm of
100+ fertilized embryos (typically FVB or FvB x B6 F1s; consult us for details and to
specify preferences). Cas9 mRNA will be provided as part of the service. All sgRNAs will
need to be purified and quantified reliably according to directions provided by the core.
Additionally, the core will simultaneously inject a DNA template for homology-directed
repair if needed for the goals of the project. Embryos that advance to 2-cell stage will be
transferred to pseudo-pregnant recipient female mice the following day, placing ~20
embryos/recipient. Users should have developed a protocol for genotyping founder pups
for editing events. Please contact the core via telephone or email to schedule a meeting

*Price: Cornell, Ithaca clients: $2,100; NY State clients: $2,800; out-of-state clients:
$3,200; non-academic clients: $5,000.
* The fees for CRISPR experiments are for services and materials, NOT for the desired end
product. We will microinject 100+ embryos (usually ~150), culture them to the 2 cell stage,
perform embryo transfers to pseudopregnant females, and deliver all pups to the PI once they
reach wean age (PI must have an animal protocol before we begin the project). We cannot
guarantee that any mice will contain the desired edited alleles, since this is subject to the
locus, the experimental design, and the quality of materials provided. Consult our web site
(transgenics.vertebrategenomics.cornell.edu ) for preferred methods of purification and other
guidelines. Quality of materials is CRUCIAL and we urge that you provide documentation of the
quality of the materials (gel pictures; method of quantification) so that potential problems can be
avoided.
