Cornell Stem Cell & Transgenic Core Facility CRISPR/Cas9 RNA Injection Description: The Core will inject purified RNAs into the pronucleus and cytoplasm of 100+ fertilized embryos (typically FVB or FvB x B6 F1s; consult us for details and to specify preferences). Cas9 mRNA will be provided as part of the service. All sgRNAs will need to be purified and quantified reliably according to directions provided by the core. Additionally, the core will simultaneously inject a DNA template for homology-directed repair if needed for the goals of the project. Embryos that advance to 2-cell stage will be transferred to pseudo-pregnant recipient female mice the following day, placing ~20 embryos/recipient.
