Such components should be avoided if possible. For example, glycine buffers and Tris buffers should not be used for samples being submitted for AAA. Additionally, alcohol-containing buffer components such as glycerol or mannitol cause degradation of certain amino acids. Proteins in PAGE gels or that have acrylamide contaminants cannot be analyzed. Proteins adsorbed onto nitrocellulose filters also cannot be analyzed. Analysis cannot be performed with samples containing detergents or antibiotics. High salt or highly acidic or basic buffers can cause instrument failure and/or interfere with hydrolysis. Proteins or peptides in PBS or PBS-like buffers (50mM phosphate, 150mM Salt, pH ~7) buffers analyze well, but higher amounts of salt or extremes of pH should be avoided. Samples that contain any of these contaminants can be analyzed, but will require a buffer exchange before AAA can occur.
