RISE STARTUP Scientific Skills Enhancement Graduate Workshop: Summer 2014 “Basic Techniques in Mammalian Tissue Culture” Workshop Leader: Dr. Steve Ontiveros Email: sjonti@nmsu.edu Dates: June 10-12 Times: 10:00am – 5:00pm Place: Foster Hall and Biology Annex Photo by Brad Shuster Objectives: This 2-day workshop is designed to familiarize graduate RISE students with the use of tissue culture systems for research purposes. The workshop will consist of one lecture and practical lab applications using a variety of mammalian cell lines. The workshop will provide basic understanding of tissue culture handling and propagation, an overview of biosecurity, biohazards and risks in working in a BSL2 facility, and expand on current transfection technologies used in tissue culture systems. Format and Scheduled Activities: DAY 1: LECTURE TOPICS 1) Working with cultured cells: BSL and safety overview 2) Cultured cell types: primary vs cell lines 3) Cultured cell types: mammalian vs teleost 3) Growth and differentiation media 4) Propagation and maintenance of cultured cells 5) Transfection: theory and techniques 6) Microscopy: phase-contrast and fluorescence DAY 1 and 2: LAB PRATICAL 1) Sterile techniques 2) Selecting and making cell culture media 3) Cell counting: what is confluency? 4) Tripsinization; sub-culture; freezing 5) Transfection 6) Imaging Maximum Capacity: 6 Supported by NMSU RISE to the Postdoctorate Program (NIH NIGMS Grant # R25GM061222) Shelley Lusetti, Graciela Unguez, Elba Serrano, (MPIs/MPDs) Contacts: Steve Ontiveros 646-5495 ; RISE Office 646-5726 1
