Induction of epileptiform convulsions in rodents, using a single injection of pentylentetrasole (PTZ) has been used in many researches especially for evaluation of antiepileptic effects of drugs or studying the neruroanatomic and neurochemical aspects of epileptic seizures. In this method the behavioral symptoms of PTZ
(50 mg/kg) are recorded and scaled according to racine method. The purpose of this study was the comparison between epileptiform behaviors after traditional method of PTZ injection with a three staged model of PTZ injection. Male mature wistar rats where divided into control (n=10) and experimental (n=10) groups. In the control group, PTZ was used in a single dose (50 mg/kg/ip) and epileptiform behaviors were recorded during one hour after injection. In the experimental group,
PTZ was injected in three separated stages (25 mg/kg each time, 15 min intervals).
If animal showed the generalized convulsions after every injection, the following injections would not proceed. If not, animal would receive the second (or third) dose of PTZ (up to 75 mg/kg). Epileptiform convulsions and behaviors after each stage of PTZ injection according to animal sensitivity will differ from partial to acute generalized seizures. The differences between this two methods are obvious and will be recorded some new behaviors that are absent in the racine scales.
So, we recommend the three stage method of PTZ injections for experimental epilepsy studies in rats.
