PREPARATION

Suspend 25 grams of the medium in one liter of distilled water. Mix well and dissolve by heating with frequent agitation.

Boil for one minute until complete dissolution. Dispense into appropriate containers and sterilize in autoclave at 121°C for 15 minutes.The prepared medium should be stored at 2-8°C. The color is amber.

The dehydrated medium should be homogeneous, free-flowing and beige in color. If there are any physical changes, discard the medium.

USES

Standard Nutrient Broth I is a medium suitable for the cultivation of fastidious bacteria. The addition of blood, ascitic fluid or serum makes it also suitable to cultivate streptococci , pneumococci and other microorganisms.

It is used for the enumeration, isolation and enrichment of bacteria, and also as high grade bases for the preparation of special culture media.

Peptone presents in the formula provide nitrogen, vitamins, minerals and amino acids essential for growth. Dextrose is the fermentable carbohydrate providing carbon and energy. Yeast extract is a source of vitamins, particularly of the Bgroup. Sodium chloride supplies essential electrolytes for transport and osmotic balance.

Inoculate and Incubate at 35 ± 2ºC during 18-24 hours.

MICROBIOLOGICAL TEST

The following results were obtained in the performance of the medium from type cultures after incubation at a temperature of 35 ± 2ºC and observed after 18- 24 hours.
