Specificity
Specific for Synacthen (1-24 ACTH) and has no cross reaction with CLIP(ACTH 17-39). Positive
with rat N-terminal ACTH peptide. Reacts <1% with insulin, KLH and BSA. Cross reacts with rat
N-terminal ACTH.

Application notes
ELISA: Use at an assay dependent dilution.
ICC: Use at an assay dependent dilution.
Reacts with solid phase ACTH either captured as part of a sandwich or adsorbed as a solid
phase antigen.
IHC-P: Use at a concentration of 1 mg/ml. Perform heat mediated antigen retrieval before
commencing with IHC staining protocol.
Not tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.
Target
Relevance
ACTH occurs in cells of the anterior pituitary and in neurons in brain. It regulates the
corticosteroid production in the adrenal cortex. Beta endorphin and Met enkephalin are
endogenous opiates. MSH (melanocyte stimulating hormone) increases the pigmentation of skin
by increasing melanin production in melanocytes.

Immunohistochemistry (Formalin/PFA-fixed
Sections were stained using an automated
paraffin-embedded sections)-ACTH antibody [57]
system DAKO Autostainer Plus , at room
(ab20358)
temperature. Sections were rehydrated and
antigen retrieved with the Dako 3-in-1 antigen
retrieval buffer citrate pH 6.0 in a DAKO PT
Link. Slides were peroxidase blocked in 3%
H2O2 in methanol for 10 minutes. They were
then blocked with Dako Protein block for 10
minutes (containing casein 0.25% in PBS)
then incubated with primary antibody for 20
minutes and detected with Dako Envision
Flex amplification kit for 30 minutes.
Colorimetric detection was completed with
diaminobenzidine for 5 minutes. Slides were
counterstained with Haematoxylin and
coverslipped under DePeX. Please note that
for manual staining we recommend to
optimize the primary antibody concentration
and incubation time (overnight incubation),
and amplification may be required.
