Just one more… • Transcription takes place where DNA is, obviously. – In which part of the cell does transcription occur? – The nucleus! Whoo! • Translation takes place in the cytoplasm with the help of a ribosome. About RNA… • RNA is another kind of nucleic acid. – Ribonucleic acid. • It is usually singlestranded, DNA is double-stranded. • Instead of having Deoxyribose, it has Ribose. • Instead of having Thymine, it has Uracil. Here’s what I got… DNA RNA Bases A, C, G, T A, C, G, U Sugar Deoxyribose Ribose Strands Two One Transcription Specifics • Really, you can think of transcription as the nucleus’s way of sending a message to the ribosome to make protein. • It just needs to transcribe the message into ribosome language first. That language is called mRNA. • mRNA stands for “messenger RNA” – makes sense, huh? Transcription • Just like in replication, the DNA is unwound, but this time by RNA Polymerase. • RNA nucleotides begin to be added to a new mRNA strand. – This time, not by DNA Polymerase but……………….. – RNA Polymerase again! (the enzyme that does it all) • Difference: Every time the mRNA molecule would have a T, a U for Uracil is placed instead. – Uracil is also a pyrimidine. Unzip DNA Helix with RNA Polymerase. – [DNA Replication uses DNA Helicase] 2. Create strand of mRNA with RNA Polymerase. – Where you would put a T for Thymine, place a U for Uracil. – [DNA Replication uses DNA Polymerase] Transcriptlate Lab • Now try number 3 on your Transcriptlate Lab. • Remember, use your original strand (printed on your worksheet) to TRANSCRIBE the mRNA and anytime you would put a T, put a U instead. • Note: DO NOT use Strand 2 here. Use Strand 1 to make a complementary Strand 3 (of mRNA), and put it on the lowest line for #3. Sense and Antisense Strands • This is confusing! • The antisense strand is the one being transcribed. – The antisense strand is sometimes called the nonsense strand. • The sense strand is the other one. It looks a lot like the mRNA but has nothing to do with it directly. – Why? ---H--Cytosine---H--- Ribose Deoxyribose Uracil ---H--Adenine---H--- hymine Deoxyribose P uanine P Ribose Guanine P P P ---H--Thymine---H--- Ribose Adenine Deoxyribose Sense Strand denine Antisense Strand Transcription Animation • [Video] Now for Translation • Translation is when the ribosome uses the mRNA strand to make a protein. – The ribosome links together amino acids based on the sequence found in the mRNA. • Quick Reminder: – Protein Monomer: Amino Acid – Protein Polymer: Polypeptide (a string of amino acids) • Quick Summary: – The two halves of the ribosome assemble around mRNA. – The ribosome “reads” the mRNA and calls over the appropriate amino acids. Why do we need translation? • Just as the word “translation” normally means converting between languages, it means almost the same thing for the cell. • It’s sort of like the nucleus speaks in DNA and the rest of the cell speaks in proteins. • RNA and the ribosomes play the role of the translator. Get those Amino Acid tables out! • Every three mRNA bases the ribosome reads is called a codon. – For example: • AUG • UUU • UCG • Each codon corresponds to a particular amino acid. – With a few exceptions. One last thing… • Protein translation always starts with the codon called AUG. To which amino acid does this correspond on your table? – By the way, methionine can also appear in the middle of the amino acid sequence. It just has to be at the front. • Protein translation always stops on one of three codons called “stop codons”: – UAA – UAG – UGA Translation Example • Here’s my mRNA sequence: – CGGAUGCUGAAGUGAGGC • Now here it is with the codons separated: – CGG | AUG | CUG | AAG | UGA | GGC • Now here it is when the amino acids linked together: – Methionine-Leucine-Lysine • Notice how I did not start at the beginning (not a start codon) and stopped at the stop codon. Let’s try it… • Try #4 on your Transcriptlate Lab. • Remember, regardless of how your strand starts, your first amino acid must be methionine. • No matter how your strand ends, stop when you reach a STOP codon. • Write your amino acids above your mRNA strand in Line 3. Last details on translation… • The amino acids that the cell strings together are originally each attached to a second kind of RNA. – tRNA (for transfer RNA) • tRNA delivers amino acids to the ribosome. – The ribosome adds the amino acids to the growing polypeptide chain (with peptide bonds) and the tRNA leaves. • The ribosome is also made of RNA (and proteins), but a different kind. – rRNA (for ribosomal RNA) • Picture next slide… tRNA and Anticodons • tRNA has a funny shape in most diagrams, but the important thing to know is that at one end is an anticodon. • An anticodon is three tRNA bases that are the opposite of the codon. Translation Steps Know these! 1. Ribosome arrives at mRNA. 2. mRNA is pulled through the ribosome. 3. Start codon (methionine) signals tRNA to start arriving. 4. One tRNA matches up to the codons on mRNA to deposit an amino acid. 5. Another tRNA comes in at the next codon and adds another amino acid to the chain. 6. Process continues until stop codon is reached. At that point the polypeptide is complete. And now it’s time for… • Codon Bingo! • Everyone needs a copy of the universal genetic code and a blank bingo card. • Write all twenty amino acids on the card. There are more than twenty spaces so some will have to be written twice (none can be written more than twice). – Don’t include stop. • I’ll give you a DNA, mRNA, or tRNA sequence – you transcribe/translate it to an amino acid. – Be sure you’re correct!
