Wash.
No cross-contamination. Genomic DNA purified from mouse tail clipping samples or water samples were
purified in adjacent wells of a 96-well plate. PCR products were amplified from 1µl of purified sample from each
well using primers for mouse IL-1β. Ten microliters of PCR product were run on a 1.5% agarose gel and
visualized by staining with ethidium bromide. Expected PCR product from mouse tail clippings for IL-1β is
approximately 1.2kb. No PCR product was detected from water samples.

System. In some cases, blood samples that are stored or shipped at elevated
temperatures contain white blood cells undergoing apoptosis. The MagneSil Blood
Genomic System purifies DNA across the broad size range seen in a typical apoptotic
ladder. Genomic DNA was isolated from blood that had been stored at the indicated
temperatures for 8 days.

1,000 base pairs were purified using the Wizard SV 96 PCR Clean-Up System on the
Beckman Coulter Biomek® 2000 robotic workstation. Panel A. Purified (P) and
unpurified (U) fragments were separated on an ethidium bromide-stained, 2% agarose
gel. Panel B. Percent recovery of purified PCR products was quantitated using a
Hitachi FMBIO® Fluorescent Scanner. Results show the mean and standard deviation
for 6 purified fragments of each size. For small PCR fragments (<500bp), optimal
recovery requires a 95% ethanol wash. For larger fragments (>500bp), optimal results
are achieved using an 80% ethanol wash.

High-quality RNA from the SV Total RNA
isolation system ensures success in
subsequent experiments such as
microarrays.
Formaldehyde gel electrophoresis of RNA. RNA was isolated from 30mg of liver
tissue following the SV Total RNA Isolation System protocol (lanes 1–6) and a
competitors protocol (lanes 7–12). Ten percent (10µl) of the final prep volume of
100µl for each sample was analyzed by electrophoresis in a 1% MOPS-EDTAformaldehyde agarose gel and visualized by including 0.1µg/µl ethidium bromide in
the sample loading buffer. Promega’s system consistently yielded an average of
100µg. The competitor’s system yielded an average of 70µg with wide variability.
