BENCH - TO - BEDSIDE TRANSLATION: 1. Discovery for the first time of: 1) Appetite Stimulating effects of Neuropeptide Y (Endocrinology 115:427, 1984), 2) Mediation by NPY Y1 receptor (Regulatory Peptides, 17:31-39, Physiol. Behav., 50:51994), 3) NPY is a physiological appetite transducer (Peptides: 9:83, 1988, PNAS, 88:1093, 1991). These discoveries have triggered development of NPY Receptor antagonists as "Appetite Suppressant" drugs by several pharmaceutical companies. 2. Demonstration for the first time that central administration of ciliary neurotrophic factor (CNTF) inhibits appetite by suppressing orexigenic NPYergic synthesis and release, with little side effects (Xu et al Endocrinology 139:466, 1998, Pu et al J. Neuroendocrinology 12:827, 2000, PNAS, 98:4279, 2001) This discovery has led to REGENERON, INC. to employ Axokine (an analog of CNTF) as a long-term weight-reducing drug. Undergoing Phase III clinical trials. 3. Demonstration for the first time that leptin gene therapy in the hypothalamus alone can ameliorate diabetes type 1 and 2and increases lifespan (Gene Therapy, 18:319, 2011).
