Preparation: Produced from sera of rabbits pre-immunized with
highly pure (>98%) recombinant human Exodus 2. Anti-human
Exodus 2 specific antibody was purified by affinity chromatography
employing immobilized human Exodus 2 matrix.

6C-kine or Exodus2 is a novel CC chemokine discovered
independently by three groups from the EST database. 6C-kine, also
named SLC (Secondary Lymphoidtissue Chemokine), and CCL21,
shows 21 33% identity to other CC chemokines. 6C-kine contains the
four conserved cysteines characteristic of β chemokines plus two
additional cysteines in its unusually long carboxyl-terminal domain.
Human 6C-kine cDNA encodes a 134 amino acid residue, highly
basic, precursor protein with a 23 amino acid residue signal peptide
that is cleaved to form the predicted 111 amino acid residue mature
protein. Mouse 6C-kine cDNA encodes a 133 amino acid residue
protein with a 23 residue signal peptide that is cleaved to generate the
110 residue mature protein. Human and mouse 6C-kine are highly
conserved, exhibiting 86% amino acid sequence identity. 6Ckine is
constitutively expressed at high levels in lymphoid tissues such as
lymph nodes, spleen and appendix. In mouse, high levels of 6Ckine
mRNA are also detected in the lung. The gene for human 6C-kine has
been localized at human chromosome 9p13 rather than chromosome
17 where the genes of many human CC chemokines are clustered.
The 6C-kine gene location is within a region of about 100 kb from
the gene for MIP3β/ ELC, another novel CC chemokine. Unlike most
CC chemokines, 6C-kine is not chemotactic for monocytes.
Recombinant mouse 6Ckine is chemotactic in vitro for thymocytes
and activated T cells. Recombinant human 6C-kine has been shown
to be chemotactic for some human T cell lines, resting PBL, and
cultured T cells expanded with PHA and IL2.

Reconstitution: Reconstitute the antibody in sterile water to a
concentration of 0.1 - 1.0 mg/ml.

Stability: The lyophilized antibody is stable for at least 2 years from
date of receipt at -20°C. The reconstituted antibody is stable for at
least two weeks at 2-8°C. Frozen aliquots are stable for at least 6
months when stored at -20°C.


Applications
Neutralization:
To yield one-half maximal inhibition [ND50] of the biological activity
of human Exodus-2 (100.0 ng/ml), a concentration of 6.0 – 8.0 g/ml
of this antibody is required.
ELISA:
To detect human Exodus-2 by direct ELISA (using 100l/well
antibody solution) a concentration of at least 0.5g/ml of this
antibody is required. This antigen affinity purified antibody, in
conjunction with compatible secondary reagents, allows the detection
of 0.2 - 0.4 ng/well of recombinant human Exodus-2.
Western Blot:
To detect human Exodus-2 by Western Blot analysis this antibody
can be used at a concentration of 0.1 - 0.2 g/ml. Used in
conjunction with compatible secondary reagents the detection limit
for recombinant hExodus-2 is 1.5 - 3.0 ng/lane, under either reducing
or non-reducing conditions.
