The effect of Xinxueshunning Capsule to resist thrombus
1. Abstract: The three dosages of Xinxueshunning Capsule, namely, large, middle and small dosage (10.75, 5.38, 2.69g crude drugs per kilogram, whose effect are 16, 8 and
4 times of that in clinic respectively ), ig, 7d, can all prolong the formation time of artery thrombus caused by electronic stimulation to the white rat. In addition, the effect will potentiate with the raise of the dosage. As for the vein thrombus caused by the postcava ligation to the white rat, the three dosages can also decrease the vein thrombus. Indication: Xinxueshunning Capsule has a good effect to resist the formation of artery and vein thrombus.
2. Purpose: observe the effect of Xinxueshunning Capsule to the formation of artery and vein thrombus.
3. Experimental materials
3.1 Animals: wistar white rat, of which both the male and the female account the half of the total number. Their weights are 180-200g and they are provided by the Medical
Institute Animal Center of province with the Acceptance Certificate: No. 03 for animal experimentation in An Hui Province. All the rats are used for experimentation after the adaptable breeding in the lab (air-conditioning temperature: 20-24).
3.2 The medicine under experimentation: Xinxueshunning Capsule is provided by
Rolling Wheel Co. Ltd, Lot number: 030662. The effect of per gram corpuscle is the same with that of 4.8g crude drugs. When using the capsule, the corpuscle should be poured into 0.5%CCMC-Na distilled water to confect the aqueous solution with three chromas, namely, 56mg/ml, 112mg/ml and 224mg/ml, for the administer drugs of three dodages.
3.3 Positive control medicine: (1) compounded Chuanxiong rhizome capsule,
370mg/corpuscle, Shandong Fenghuang Compounder, Lot number: 020627. When using the capsule, the corpuscle should be confected with 0.5%CCMC-Na distilled water with the chroma of 59.2mg/ml. (2) Aspirin tablet, 100mg/tablet, produced by
Barfer, Lot number: xj374. When being used, aspirin should be comminuted in mortar and be confected with 0.5%CCMC-Na distilled water with the chroma of 4mg/ml; (3)
Warfarin tablet, 2.5mg/tablet, Shanghai Jiufu Medical Co. Ltd, Lot number: 030421.
When using the tablet, Warfarin should be comminuted and be confected with
0.5%CMC-Na distilled water with the chroma of 0.033mg/ml.
3.4 Instrument: BT-3 Thrombus-formation-Mensurating Instrument of intestine thrombus formation, the prodrct of Cardiovascular Institute, Baotou Medical College.
4. Methods and results
4.1 The effect of Xinxueshunning Capsule to the carotid thrombus caused by the electronic stimulation
48 wistar white rats of which both male and female account the half of the total number are chosen and divided into 6 groups with 8 white rats per group. 0.5%CCMC-Na distilled water is given to the normal control; positive medicine of aspirin group 40mg/kg
(the effect is 8 times of that in clinic), compounded Chuanxiong rhizome group 592mg/kg
(the effect is 8 times of that in clinic); Xinxueshunning Capsule with three dosages of large, middle and small (10.75, 5.38, 2.69g crude drugs/kg, whose effect is 16,8,4 times of that

in clinic). The administration volume of every group is 10ml/kg, one time of ig every day lasting 7 days. In the night before the experimentation, feeding is forbidden and watering is permitted. One hour after the last administration, the rats are hocused by chloral hydrate (0.35g/kg). The cervix skin is dissected and the right carotid is separated. The plastic film is put under the separated carotid to avoid the contact between the stimulating electrode and the surrounding issues. The stimulating electrode is put under the proximal end of the artery and the temperature measuring head which is connected with the apparatus is put under the distal end. 1.5mA direct current from the stimulating electrode stimulates the vascular wall for two minutes and injuries it. With the gradual formation of the thrombus in the blood vessel, blood stream is gradually interdicted and the temperature of the proximal end of artery gradually declines. When the blood stream is completed interdicted and the temperature declines suddenly, the apparatus will give an alarm and show the blocking time (OT), namely, forming time of thrombus. The result indicates: three-dosage groups of Xinxueshunning Capsule can prolong the forming time of artery thrombus and the effect will potentiate with the raise of the dosage. The dependency to the dosage indicates that the capsule has a good effect to resist the formation of artery thrombus; so does the positive medicine.

4.2 The effect of Xinxueshunning Capsule to the formation of vein thrombus
48 wistar white rats of which both male and female account the half of the total number are chosen and divided into 6 groups with 8 white rats per group. 0.5%CCMC-Na distilled water is given to the normal control; three dosages of Xinxueshunning Capsule,

namely, large, middle and small (10.75, 5.38, 2.69g crude drugs respectively, whose effect is 4, 8 and 16 times of that in clinic); positive control: compounded Danshen Tablet group
300mg/kg (whose effect is 8 times of that in clinic), Warfarin group 0.33mg/kg (whose effect is 4 times of that in clinic). The administration volume of every group is 10ml/kg, one time of ig every day lasting 7 days. In the night before the experimentation, feeding is forbidden and watering is permitted. One hour after the last administration, the rats are hocused by chloral hydrate (0.35g/kg). The abdominal cavity of white rat is opened from white line. The postcava is separated from the surrounding issues by the glass needle and is ligated with seam under the left vena renales. The ligating positions of the white rats are kept concordant and the abdominal cavities are stitched. Two hours after the ligation, the abdominal cavity is opened and the vessel 2cm below the ligating position is held by haemostat. The residual blood in the vessel is aspirated by needle tubing, the vein wall is laid open carefully and the thrombus is brought out on the weighing paper. After the floating blood around the thrombus is imbibed by filter paper, the thrombus is weighed by electronic analytic balance.
The result indicates: Three dosages of Xinxueshunning Capsule can all decrease the formation of vein thrombus; positive control is also effective in resisting the formation of vein thrombus and the effect of Warfarin is the strongest. (see sheet 2)

5. Conclusion
The three dosages of Xinxueshunning Capsule, namely, large, middle and small
(10.75, 5.38, 2.69g crude drugs, whose effect are 16, 8, 4 times of that in clinic) can

prolong the forming time of artery thrombus caused by the electronic stimulation to the white rat. In addition, the effect will potentiate with the raise of the dosage. As for the vein thrombus caused by the ligation of postcava to the white rat, all the three dosages can decrease the formation of vein thrombus obviously. Indication: Xinxueshunning Capsule has a good effect to resist the formation of thrombus.
